目的分析胃腺癌BGC823细胞Zbtb7A高表达前后差异基因的表达。方法应用22K人基因组寡核苷酸芯片,筛选BGC823细胞高表达Zbtb7A前后差异表达的基因,采用RT-PCR验证部分差异基因的表达。结果发现Zbtb7A高表达前后BGC823细胞有显著性表达差异的基因共312个,其中上调基因205个,下调基因107个。经RT-PCR验证部分差异基因的表达与芯片结果一致。结论Zbtb7A高表达前后胃腺癌BGC823细胞部分功能基因存在着显著的表达差异,多数差异表达的基因参与细胞黏附、细胞骨架形成以及细胞代谢等过程。
Objective To analyze the expressions of diEerential genes in gastric adenocarcinoma BGC823cells before and after ZbtbTA overexpression. Methods The different expressive genes in BGC823 cells before and after ZbtbTA overexpression were screened by 22K human genome oligonucleotide array. Then RT-PCR was used to verify the expressions of some differential genes. Results A total of 312 genes was screened out, the expression level of which was significantly changed after ZhtbTA overexpression. Among these genes,205 genes were upregulated and 107 genes were downregulated. Moreover, the result of verification test was coincident with that of gene chip. Conclusion Significant differences are found in the expressions of some functional genes in BGC823 cells before and after Zbtb7A overexpression. Most of these genes are involVed in the development of cell adhesion, cytoskeleton and metabolism
