目的观察大鼠椎间盘脱出模型的脊髓血流量及其电针效应,为兽医临床犬椎间盘病病理和针灸作用机理研究积累资料。方法通过外科手术将硅胶片填充于大鼠第13胸椎(T13)脊髓腹侧位制作胸腰段椎间盘脱出模型,通过激光散斑血流系统实时监测造模后第一腰椎(L1)脊髓背侧血流量及电针刺激双侧足三里穴和趾间穴对其的影响。结果以硅胶片填充于大鼠T13的脊髓腹侧位可成功制作出类似犬胸腰段椎间盘脱出的后肢瘫痪模型,且重复性好;模型组大鼠在压迫后,L1段的脊髓背侧血流量呈极显著下降(P〈0.01),电针双侧足三里穴和趾间穴可显著升高血流量(P〈0.05),电针结束后约10 min其血流量逐渐恢复至电针前水平;电针组大鼠电针治疗14 d后,脊髓背侧血流量极显著高于对照组,且运动功能评分显示电针组大鼠的后肢运动功能显著改善。结论通过外科手术将硅胶片填充于大鼠T13脊髓腹侧位可成功制作胸腰段椎间盘脱出模型;压迫后L1段脊髓血流量显著下降;电针治疗后脊髓血流量极显著升高,且瘫痪大鼠运动机能明显好转,这可能与电针改善脊髓血流量继而减轻脊髓损伤并促进损伤恢复或促进代偿机制有关。
Objective To observe the changes of spinal cord blood flow(SCBF) in rat models of intervertebral disc extrusion and the effects of electroacupuncture(EA),and to get better understanding the pathology of canine intervertebral disc disease and the mechanism of acupuncture treatment in veterinary clinics.Methods Thirty seven male adult Sprague Dawley rats were randomly divided into 3 groups: the normal control group(9 rats),model control group(14 rats) and electroacupuncture-treated model group(14 rats).First,the rat model of intervertebral disc extrusion was established by filling the thoracic spinal canal at the level of 13th(T13) thoracic vertebra with a silicone sheet to compress the spinal cord from the ventral side.The SCBF at the level of the first lumbar vertebrae(L1) and the effects of electroacupuncture in real time were monitored in the rat models by a laser speckle blood flow system.The selected acupoints in this experiment were Zusanli and Zhijian.Results This type of rat model of intervertebral disc extrusion could successfully simulate canine intervertebral disc extrusion,and it had a good repeatability.When the model was completed,the SCBF at the level of L1 decreased significantly(P0.01).During EA treatment at the bilateral acupoints of Zusanli and Zhijian,the SCBF in EA group increased significantly(P0.05).After EA treatment,the SCBF was slowly increased to that of before EA in about 10 minutes.At 14 days after EA treatment,the SCBF in the EA group was significantly higher than that of the control group,and the score of motor function showed that the hindlimb motor function in the EA group was significantly improved as compared with that of the control group.Conclusions A rat model of intervertebral disc extrusion has been successfully developed by filling the spinal canal with a silicone sheet at the level of 13th thoracic vertebra to compress the spinal cord from the ventral side,and the SCBF at the level of L1 decreased significantly after the compression.Th
