目的利用9.4T磁共振频谱仪离体检测3种乳腺癌细胞的水溶性代谢物频谱特征。方法培养获取ZR-75-1、T47-D及SK-BR-3乳腺癌细胞,用高氯酸提取水溶性代谢物,以9.4T高场磁共振仪检测所提取水溶性代谢物的氢质子频谱,定量计算谱线中相应代谢物的浓度并分析其特征。结果 ZR-75-1、T47-D及SK-BR-3乳腺癌细胞具有3种不同代谢物特征;3种乳腺癌细胞在乳酸及胆碱处均出现波峰,且T47-D的乳酸及胆碱浓度高于ZR-75-1和SK-BR-3。结论利用9.4T磁共振频谱仪能获取3种乳腺癌细胞的代谢信息;T47-D乳腺癌细胞内乳酸及胆碱代谢高于ZR-75-1和SK-BR-3。
Objective To explore the features of spectrum of water soluble metabolites of 3 kinds of breast cancer ceils in vitro with 9.4T 1 H-MRS. Methods Three kinds of human breast carcinoma ceils (ZR-75-1, T47-D, SK-BR 3) were cultured and collected. The metabolites were extracted with perchloric acid and prepared for MRS analysis. ^1H spectra were acquired with 9.4T high resolution MRS, and the concentrations of main metabolites were quantitatively calculated. Results ZR-75-1, T47-D and SK-BR-3 breast cancer ceils had characteristics of three different metabolites. The metabolic peak of lactate and choline were observed in all 3 kinds of breast cancer cells. The concentration of choline-compounds and lactate in T47-D were higher than those of ZR-75-1 and SK-BR-3. Conclusion The metabolic profiles of three subtypes of human breast carcinoma cells can be reliably detected and quantified with 9.4T MRS. The metabolic of lactate and choline-compounds of T47-D cell are higher than those of ZR-75-1 and SK-BR-3.
