目的探讨巢蛋白(nestin)和缺氧诱导因子-1α(HIF-1α)在人脑胶质瘤组织中的表达及其在皮质瘤血管生成中所起的作用。方法应用免疫组化两步法检测53例不同级别胶质瘤和5例正常胎脑组织中nes—tin和HIF-1α的表达。结果nestin在低级别和高级别胶质瘤的平均光密度值(A)分别为0.2463±0.0602和0.6438±0.0901,差异有显著性(P〈0.05);HIF-1α在低级别和高级别胶质瘤的平均光密度值(A)分别为0.1334±0.0298和0.2746±0.0904,差异有显著性(P〈0.05)。结论胶质瘤中,nestin在新生微血管内皮细胞和HIF-1α仅在肿瘤细胞的表达与胶质瘤的病理分级有关,级别越高表达越强,而且两者呈正相关,提示可将nestin和,HIF-1α作为一种判断胶质瘤新生血管增生、恶性程度的参考依据。
Objective To investigate the expression of nestin and HIF -1α in human glioma and their roles in the formation of blood vessels. Methods Immunohistochemistry was used to detect the expression of nestin and HIF -1α in 53 cases of human gliomas of different grades and 5 cases of normal fetus brains. Results The average optical density (A) of nestin in low grade and high grade of gliomas was O. 246 3 ± 0. 060 2 and 0. 643 8 ± 0. 090 1, respectively, with significant difference between the two groups ( P 〈 0.05 ). The average density ( A ) of HIF - -1α in low grade and high grade of gliomas was O. 133 4 ±0. 029 8 and O. 274 6 ±0. 090 4, respectively, with significant difference between the two groups ( P 〈 0.05 ). Conclusion The expression of nestin in newborn vascular endothelial cells and the expression of HIF -1α in glioma cells are associated with the grades of gliomas. Expression of nestin and HIF -1α can be used to evaluate vascular proliferation and malignancy of gliomas.
