目的:研究当归及其主要成分阿魏酸钠(SF)对促进脑缺血再灌注后神经功能恢复的作用,并将两者进行比较。方法:雄性Wistar大鼠60只,改良线栓法制作大脑局灶性脑缺血再灌注模型后(n=45),分别分为当归组、SF组和对照组,每组15只,假手术组(n=15)线栓仅插入1cm,各组动物分别取2,7,14d3个时间点,观察神经功能恢复、脑梗死体积、脑含水量。结果:当归组Bederson评分在术后第7天开始优于对照组(P〈0.05),SF组在第2,7天均优于对照组(P〈0.05)。SF组横木行走测试评分在各时间点均高于对照组(P〈0.05),当归组与对照组相比无显著性差异。SF组足失误测试评分在术后2,14d低于对照组(P〈0.05),当归组与对照组相比无显著性差异。SF组脑梗死体积在各时间点均明显小于对照组,且在14d小于当归组(P〈0.05),当归组只在术后2,14d小于对照组(P〈0.05)。除SF组外,各组脑含水量均呈逐渐上升趋势,SF组在术后2d高于其它各组,术后14d低于其它各组(P〈0.05)。当归组在术后7,14d低于对照组(P〈0.05)。结论:当归和SF都能改善脑缺血再灌注后的神经功能,减小脑梗死体积,减轻脑水肿,但在促进神经功能恢复方面,SF可能发挥早期作用,减小脑梗死体积和减轻脑水肿的功能的作用强于当归,SF可能是当归治疗脑缺血再灌注损伤的主要有效成份。
Objective. To compare the effects on neurological function after reperfusion of focal cerebral ischemia between angelica sinensis and sodium ferulate (SF), the main component of angelica sinensis. Methods: There were 60 male Wistar rats involved in the study and 45 were subjected to focal cerebral ischemia and divided into three groups equally as angelica group, SF group and control group,and the other 15 rats were only performed with sham operation and the threads were only inserted 1 cm deep into the carotid arteries. The effects on neurological function, infarct volume and brain water content were investigated at different time points in both angelica group and SF group after reperfusion of focal cerebral ischemia. Results: Both angelica and SF could significantly enhance the recovery of the neurological function. There were no differences among the groups at 14 d after surgery. SF group got higher scores than control group at each time point in beam walking test and at 2 and 14 d in foot-fault test (all P〈0.05). Angelica group had no differences compared with control group in both tests. Infarct volume of angelica group was significantly decreased at 2 d and 14 d, but SF group were significantly decreased at each time point compared with control group. Brain water contents in all groups were gradually increased except in SF group, and in angelica group were lower than in control group at 7 d and 14 d. Conclusion. Both angelica and SF can promote neurological function, and diminish infarct volume and cerebral edema. SF seems to enhance the neurological function in early period and has better effects on diminishing infarct volume and cerebral edema than angelica sinenesis does.
