目的:探讨凝血酶对食管癌细胞EC109细胞迁移的影响。方法:EC109分别接种于96孔培养板各孔,贴壁后划痕,分别加入不同浓度的凝血酶,作用时间12、24、48、72h。同时设置水蛭素组、凝血酶+水蛭素组及无药组。拍照并测量计算细胞迁移距离。结果:凝血酶组各浓度在不同作用时间的迁移距离与水蛭素组、凝血酶+水蛭素组和无药组比有统计学意义(P〈0.05),水蛭素组、凝血酶+水蛭素组与无药组比无统计学意义,凝血酶组各浓度间比无统计学意义。结论:凝血酶可增强EC109细胞迁移能力。
Objective:To investigate the effect of thrombin on EC109 cells migration in esophageal carcinoma. Me-thods:EC109 cells were cultured in a 96-well culture plate. The cell monolayer was scraped in a straight line to create a "scratch" with a 10 μL pipet tip. The EC109 cells were cultured with thrombin,hirudin,thrombin+hirudin in different concentrations. After 12,24,48,72 hours,the image of the scratch was acquired,then the distances of EC109 cells migration were measured and calculated. Results:Following 12,24,48,72 hours incubation with thrombin,the EC109 cells migration was obviously better than of groups(P〈0.01). The EC109 cells migration among different concentrations of thrombin had no statistical significance. Conclusion:The thrombin may enhance EC109 cells ceumigration.
