目的:探讨低抗凝肝素对胎牛血清培养的大鼠肝星状细胞生长的影响。方法:肝星状细胞经体积分数为0.4%胎牛血清/DMEM培养液同步化48h后分为对照组和实验组,对照组以体积分数为10%胎牛血清处理,实验组分别以体积分数为10%胎牛血清和不同质量浓度(8g/L,40g/L,200g/L,1000g/L,5000g/L)低抗凝肝素处理,应用MTT法测定其对肝星状细胞生长的影响。结果:低抗凝肝素(40~5000g/L)对肝星状细胞的增殖有明显抑制作用(P〈0.05)。在不同的作用时间,低抗凝肝素(200g/L)均对肝星状细胞生长有明显抑制作用,且在其作用48h后肝星状细胞生长速度明显减慢。结论:低抗凝肝素对大鼠肝星状细胞生长具有抑制作用。
Aim : To explore the effect of low anticoagulative activity heparin on the growth of rat hepatic stellate cells (HSCs). Methods:Rat HSCs were treated with 0.4% fetal calf serum (FCS)/DMEM for 48 hours and divided into the control group and experiment groups. HSCs in the control group were treated with 10% FCS, and HSCs in the experiment groups were treated with 10% FCS and low anticoagulative activity heparin with different concentrations ( 8 g/L, 40 g/L, 200 g/L, 1 000 g/L, 5 000 g/L). MTT reduction assay was used to evaluate the effect of low anticoagulative activity heparin on the growth of rat HSCs. Results: Low anticoagulative activity heparin with different concentrations ( 40 - 5 000 g/ L) all inhibited the growth of HSCs significantly. At different acting time points, low anticoagulative activity heparin (200 g/L) could inhibit the growth of HSCs significantly, especially after 48 hours. Conclusion: Low anticoagulative activity heparin can inhibit the growth of rat HSCs.
