目的探讨饮水摄入三氯乙烯(trichloroethylene,TCE)对小鼠Th17细胞的影响。方法将48只6周龄雌性BALB/c小鼠按随机数字表法分为空白对照组、溶剂对照组、2.5mg/ml TCE组和5.0mg/ml TCE组,每组12只,染毒组经饮水摄入TCE;每组分别于暴露第14、28、56、84天各处死3只小鼠,取其脾脏及外周血,流式细胞术检测CD4+T细胞中Th17细胞比例,实时荧光定量PCR测定维甲酸相关孤儿核受体(retinoid—related orphan receptors,RORTγt)mRNA表达水平;ELISA法测定外周血血清白细胞介素-17(IL-17)、白细胞介素-6(IL-6)和转化生长因子-β(TGF—β)含量。结果2.5、5.0mg/ml TCE染毒组小鼠的脾脏CD4+T细胞中Th17细胞比例第14天时分别为(3.46±0.32)%和(5.45±0.45)%,第84天时分别为(3.47±0.33)%和(4.10±0.39)%,均较同期溶剂对照组[分别为(2.15±0.20)%、(2.16±0.35)%]升高且差异有统计学意义(P值均〈0.01);其RORγt表达水平第14天时分别为1.870±0.084和1.965±0.060,第56天时分别为1.998±0.079和2.028±0.073,均较同期溶剂对照组(1.77±0.04、1.75±0.09)上调且差异有统计学意义(P值均〈0.05);其血清IL-17含量第14天时分别为(32.28±5.38)和(34.47±5.02)pg/ml,第28天时分别为(34.87±5.48)和(41.94±6.19)pg/ml,均较同期溶剂对照组升高[分别为(21.57±5.23)、(22.11±5.11)pg/ml]且差异有统计学意义(P值均〈0.05);其血清IL-6含量第14天时分别为(43.07±6.71)和(47.86±8.52)pg/ml,第56天时分别为(41.32±7.04)和(46.74±9.33)ps/ml,均较同期溶剂对照组[分别为(7.56±7.71)和(28.26±7.22)pg/ml]升高且差异有统计学意义(P值均〈0.05);其血清TGF—β含量第14天时分别为(17.48±3.06)和(18.93±3?
Objective This study aimed to investigate the effect of trichloroethylene (TCE) intake via drinking water on Th17 cells in mice. Methods Forty eight six weeks old female BALB/c mice were divided into blank control,vehicle control,2. 5 mg/ml TCE and 5.0 mg/ml TCE groups by random number table( 12 mice each group) , and exposed to TCE by drinking water. On the 14th , 28th, 56th, 84th days, blood were collected and assayed for IL-17 ,IL-6, and TGF-β concentration in serum through ELISA. Animals were killed and spleen biopsies were taken sterilely. The proportion of Tb17 cells among CD4 + T cells and RORγt mRNA expression level in spleen were measured by FCM and real-time PCR.
