【目的】为了验证COI1基因在葡萄抗病防御机制中的作用,【方法】在前期通过抑制消减杂交技术(SSH)获得中国野生葡萄毛葡萄株系‘商-24’COI1基因EST序列的基础上,采用RT-PCR从毛葡萄‘商-24’中分离到COI1基因cDNA序列,并通过实时定量PCR和半定量PCR分析了COI1基因的表达特异性,以期验证其在葡萄抗病防御机制中的作用。【结果】序列分析表明,VqCOI1基因长2 455 bp,编码598个氨基酸残基,相对分子量为67.87 ku,等电点为5.85。同源性分析表明,它与已克隆的大豆(Glycine max)COI1基因(GenBank登录号:AAZ66745.1)相似性最高为79%。实时定量PCR技术用来检测VqCOI1基因在受到病原菌侵染和激素处理时在抗病品种‘商-24’中的表达情况,结果表明,在白粉菌诱导初期,抗病的毛葡萄株系‘商-24’VqCOI1基因表达量升高,这可能是因为它参与了病原菌诱导的JA信号调节途径。水杨酸(SA)、茉莉酸甲酯(MeJA)、乙烯(Eth)处理表明VqCOI1基因表达受激素分子调节,这些信号分子可能交叉调节这一基因的表达。在嫩叶、嫩茎、花、果皮、卷须等不同葡萄组织中的表达表明Vq-COI1基因的表达具有空间差异性。【结论】这些结果暗示着VqCOI1的表达响应病原菌侵染并受生物胁迫相关激素的调控,很有可能在葡萄防御病原菌侵染的机制中发挥重要作用。
【Objective】 The objective of the study is to verify the role of COI1 gene in grape disease resistance.【Method】Based on a EST sequence of COI1 gene in the subtractive suppression hybridization(SSH) cDNA library from young leaves of Chinese wild Vitis quinquangularis clone 'shang-24' inoculated with Uncinula necator(Schw.) Burr,we isolated the cDNA sequence of that COI1 gene using the RT-PCR technology,which was designated as VqCOI1.Then we studied its expression patterns by Semi-Quantitative RT-PCR and Quantitative RT-PCR,on the purpose of verifying its roles in grape disease resistance.【Result】The sequence analysis indicated that the length of VqCOI1 was 2 455 bp,which encodes 598 amino acids,with a predicted molecular weight of 67.87 ku and the isoelectric point of 5.85.Multiple Alignment of deduced amino acid sequence showed that it has the highest sequence similarity,approximately 79% similarity with other COI1 in Glycine max(GenBank acc.no: AAZ66745.1).The qRT-PCR showed that the expression of VqCOI1 was induced and increased in the early stages when inoculated with U.nector in the resistant line 'Shang-24',which suggested that VqCOI1 might be involved in pathogen-induced JA signal regulatory pathways.VqCOI1 was also regulated by Ethylene(Eth),Methyl jasmonate(MeJA) and Salicylic acid(SA),and these signaling molecules may cross-regulate the expression of this gene.VqCOI1 was also expressed differentially in leaves,stems,flowers,pericarps and tendrils.【Conclusion】 These results revealed that VqCOI1 was a pathogen-responsive gene and it was regulated by certain biotic stress-related hormones,indicating it may play an important role in grape pathogen resistance.
