目的在真核小鼠胚胎成纤维细胞(MEF)内,进行不同浓度或不同时间的氯化镍(Ni Cl2)刺激,观察超氧化物歧化酶2(SOD2)的变化,对其可能机制进行探讨。方法培养MEF系,制备Ni Cl2溶液进行处理,利用Western blotting观察不同浓度或不同时间的Ni Cl2刺激后SOD2的变化。随后判断其可能的机制。结果不同浓度的Ni Cl2刺激MEF 12 h后,SOD2的表达总体上有下降趋势,磷酸化蛋白激酶B的表达总体上有上升趋势,而磷酸化信号转导和转录激活子(p-Stat3)的表达总体上有下降趋势;相同浓度的Ni Cl2刺激MEF不同时间后,SOD2和p-Stat3的表达总体上有下降趋势;p-Stat3与SOD2的总体变化趋势一致。结论 SOD2在受到Ni Cl2的刺激作用后,总体上呈现下降趋势;p-Stat3可能是作为转录因子,在转录水平上进行了SOD2的调控。
Objective In MEF eukaryotic cells,it was stimulated with NiCl2 with different concentrations and times. It is investigated the change of SOD2 and its potential mechanisms. Methods Cultivating MEF cells line and preparing Nicl2 solu-tions, it was observed the change of SOD2 stimulated by NiCl2 of different concentrations and times by Western blotting and then judged the potential mechanism. Results After stimulating MEF for continuous 12 hours with different-concentration NiCl2,the expression of SOD2 was orientated to declining and p-Stat3 and p-Akt rising respectively in general. After stimulating MEF for dif-ferent time period with the same-concentration NiCl2,the expressions of SOD2 and p-Stat3 were both orientated to decline and ris-ing respectively generally. The overall change of SOD2 and p-Stat3 were in consistent. Conclusion Stimulated by NiCl2,SOD2 showed a trend of decline,p-Stat3, probably as the transcription factor,adjusted SOD2 on the transcriptional level.
