研究L-谷氨酰胺(L—glutamine,GLN),Nα-乙酰-L-谷氨酰胺(Nα-acetyl—L—glutamine,NAG),L-精氨酸(L—arginine,ARG),N—α-乙酰-L-精氨酸(N—alpha—acetyl—L—arginine,NAA)、瓜氨酸(L—citrulline,CIT),α-酮戌二酸(Alpha—ketoglutaric acid,AKG)和L-脯氨酸(L—Proline,PRO)对猪小肠上皮细胞(Intestinal porcine epithelial cells,IPEC—1)细胞活力与生长的影响,比较这几种氨基酸及其衍生物对IPEC—1细胞生长的作用效果,在IPEC—1定制培养液中分别添加不同浓度的氨基酸或其衍生物培养4d。用CCK8(cell counting kit 8)法检测细胞的活力,并用细胞计数法检测这些氨基酸或其衍生物对细胞生长的影响试验的整个4d,GLN组IPEC—1细胞的数量(P〈0.05)和OD值(P〈0.05)均显著高于其他组,在试验的第4d,NAG、AKG、CIT、NAA和ARG组细胞数量均高于对照组(P〈0.05或0.01),培养液中添加PRO对IPEC—1细胞的生长无明显影响。①在这几种氨基酸及其衍生物中,GLN促进IPEC—1细胞生长的作用效果最好,②NAG、AKG、CIT、NAA或ARG均能提高第4d IPEC—1细胞的数量。
This study was conducted to investigate the effects of L-glutamine (GLN), Nα-acetyl-L-glu- tamine (NAG), L-arginine (ARG), N-alpha-acetyl-L-arginine (NAA), L-citrulline (CIT), Alpha-keto-glularic acid (AKG) and L-Proline (PRO) on the cellular vitality and proliferation of intestinal porcine epilhelial cells (IPEC-1). The IPEC-1 cells were incubated with the corresponding concentraliuns of amino acids or their derivatives for d days. The cellular vitality was measured by CCK8 (cell counting kil 8), and the cell numbers were recorded. The IPEC-1 cell numbers and OD value in GLN group were higher than the other groups (P〈0.05).The IPEC-1 cell numbers in NAG, AKG, CIT, NAA and ARG groups were higher than the control group (P〈0.05 or 0.01). There were no significant aherations of IPEC-1 cell numbers hy PRO supplementation. ①GLN is the most effective amino acid in increas-ing the IPEC-1 eell numbers and vitality. ②NAG,AKG, CIT, NAA and ARG can promote the cell growth on the fourth day 4 of the trial.
