目的研究电针对MCAO大鼠脑组织小胶质细胞TLR4及NF-κB蛋白表达的影响。方法建立MCAO大鼠模型,分别电针内关、曲池后进行神经体征评分,HE染色检测神经元坏死率,双重免疫荧光染色检测TLR4、NF-κB p65蛋白在小胶质细胞的表达。结果模型组与正常组比较,大鼠神经体征评分、脑海马神经元坏死率、脑组织TLR4、NF-κB p65蛋白表达均明显升高,差异有统计学意义(P〈0.01);内关组、曲池组与模型组比较,神经体征评分、脑海马神经元坏死率、TLR4、NF-κB p65蛋白表达明显降低,差异有统计学意义(P〈0.05);曲池组与地机组比较,脑海马神经元坏死率明显降低,差异有统计学意义(P〈0.05);内关组与地机组比较,大鼠脑组织小胶质细胞TLR4蛋白表达明显降低,差异有统计学意义(P〈0.05)。结论电针内关、曲池能改善MCAO大鼠的神经功能损伤,抑制TLR4及NF-κB蛋白在小胶质细胞的表达,提示电针能够通过影响小胶质细胞TLR4/NF-κB信号通路的活化从而发挥脑保护作用。
Objective To investigate the effects of electroacupuncture on the protein expression of TLR4 and NF - KB p65 in microglia cells in cerebral of middle cerebral artery occlusion ( MCAO ) Rats. Methods After electroacupuncture on Neiguan ( PC6 ) and Quchi ( LII 1 ) respectively, we observed neurologi- cal syndrome score of MCAO rats. The death rate of neuron were measured by HE dyeing, and the protein ex- pression of TLR4 and NF - KB p65 in microglia cells were detected by doubling immunofluorescent staining. Results Compared to sham - operation control group, neurological syndrome score, death rate of neuron and the protein expression of TLR4 and NF - KB p65 in mieroglia cells were increased in model group (P 〈 0.01 ). Compared to model group, neurological syndrome score, death rate of neuron, the protein expression of TLR4 and NF - KB p65 in microglia cells were decreased in Neiguan and Quchi group( P 〈 0.05 ). Compared to Diji group, the death rate of neuron were decreased in Quehi group( P 〈 O. 05 ), and the protein expression of TLR4 in microglia cells were decreased in Neiguan group ( P 〈 0.05 ). Conclusion After eleetroaeupunc- ture on Neiguan and Quehi, the damage of cerebral nerve function of MCAO rats are improved, and the ex- pression of TLR4 and NF - KB p65 in mieroglia cells are inhibited. It indicates that through effects on the ac- tivation of TLR4/NF - KB signal pathway in microglia cells, electroacupuncture can play a protective role on cerebral ischemia.
