本实验建立了饲料中喹乙醇的固相萃取和高效液相色谱法定量检测及质谱确证的方法。饲料经5%甲醇溶液提取,离心,过滤,移取2mL滤液注入已活化的3mL HLB固相萃取小柱,分别用3mL 0.02mol/L的盐酸溶液和5%甲醇溶液淋洗,用2mL 40%甲醇溶液洗脱,洗脱液经微孔滤膜过滤后上机定量检测,同时滤液进行质谱确证。试验结果表明,喹乙醇浓度为0~100μg/mL时具有良好的线性关系;喹乙醇回收率为86%~100%。
A high performance liquid chromatography (HPLC) and liquid chromatography-mass spectrometry (LCMS) method for determination of olaquindox in feeds were developed.Olaquindox in feeds were extracted with 5 % methanol solution and further clean up with HLB solid phase extraction eartridge.LC-MS was in an electrospary positive ion mode.The results showed that the average recoveries were 86 % - 100 %.
