目的:荧光探针法研究石榴皮鞣质对溴化乙锭(EB)引起的大鼠肝细胞DNA损伤的保护作用。方法:培养大鼠肝细胞,抽提DNA,建立EB-DNA体系,研究3种样品溶液(石榴皮鞣质提取物,鞣花酸-没食子酸混合溶液,以及维生素C)与DNA分子的作用。根据作用常数的大小讨论了3种溶液及其各自的浓度对DNA的保护作用。并初步推断作用机制。结果:3种溶液对体系的荧光抑制率都随浓度增加而增加,其中石榴皮鞣质提取液组的抑制率可达到97%以上,鞣花酸-没食子酸组具有饱和性,维生素C组的抑制率最低。结论:石榴皮鞣质提取液与DNA分子可发生显著作用,对EB引起的大鼠肝细胞受损DNA有保护作用。
OBJECTIVE The protection of DNA by pomegranate pericarps (Punica granatum. L) tannins extract was studied by using fluorescence probe EB. METHODS Rat hepatocytes were cultivated, DNA was extracted, EB-DNA system was investigated the influence of three samples (pomegranate pericarps tannins, ellagic acid galic acid , VitC)on the fluorescence intensity was investigated with the fluorescent probe of EB. The protection of DNA and the mechanism were discussed. RESULTS The inhibition rate of relative fluorescence intensity increased with increasing concentration of the three antioxidants. Nevertheless, one treated with tannins of pomegranate pericarps was not saturated when the concentration varied, even above 97 percent,one treated with ellagic acid galic acid had a saturation character and the other treated with VitC had the lowest inhibtion rate. CONCLUSION Tannins from pomegranate pericarps can protect rat hepatocytes against DNA damage induced by EB.
