目的为探讨清醒状态下尾静脉多次反复给药造模方法及舒芬太尼反复给药对大鼠记忆损伤的影响。方法取实验用2-3月龄大雄性SD大鼠80只。实验组分别静脉注射舒芬太尼高浓度25μg/kg(s1组,n=27),舒芬太尼低浓度12.5μg/kg(S2组,n=21)。空白对照组舒芬太尼0μg/kg(C组,n=26),给予等体积的Ns。实验组每天注射一次,至7、14d时观察大鼠水迷宫测试。记录水迷宫的测试结果,与空白对照组进行分析对照得出结论。结果(1)舒芬太尼s1组大鼠7、14d时,与空白对照组大鼠比较,逃逸潜伏期时间明显延长(P〈0.05)、14d时跨平台次数与空白对照组比较明显减少(P〈0.01);(2)舒芬太尼s1组与s2组大鼠比较,大鼠7d、14d时逃逸潜伏期均时间明显延长(P〈0.05)、14d时跨平台次数明显减少(P〈0.01)。结论大鼠尾静脉多次穿刺给药方法的改进,可提高穿刺成功率。舒芬太尼高浓度尾静脉注射2W,可引起SD大鼠的空间学习记忆能力损伤,呈时间和剂量依赖性。
Objective To improve the method of multi-time tail vein injection of rats in waking state and to inveti- gate influence of sufentanil to the injury on spatial learning and memory. Methods 80 healthy male Sprague-Dawley rats were divided into three groups randomly, including sufentanil 25μg/kg ( S1, n = 27 ), sufentanil 12.5 μg/kg ( S2, n = 27 ) and control group. Control group(C,n =26)were injected the same volume of NSat the end of the7, 14 days. To observe changes of behavior. Compared with normal group and record the result of the water maze of rats of every group. Results ( 1 ) Compared with the control group rats, at 7 and 14 day, the escape latency time was significantly longer in Sufentanil 25 μg/kg group rats( P 〈 0.05), compared with the blank group cross-platform times was decreased significantly at 14 days (P 〈 0.01 ). ( 2 ) At 7 and 14 day, rats escape latency time of sufentanil 25 p.g/kg group prolonged significantly compared with sufentanil 12.5 μg/kg rats, ( P 〈 0.05 ), cross-platform times compared with sufentanil 12.5 μg/kg group decreased signifi- cantly at 14 day(P 〈 0.01 ). Conclusion Tail vein injection of 25 μg/kg sufentanfl for 2 weeks in SD rats has a signifi- cant injury on spatial learning and memory.
