目的观察补肝健腰方对腰椎间盘退变大鼠髓核组织肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)表达的干预作用,探讨补肝健腰方防治腰椎间盘退变的疗效机制。方法 40只10月龄SD大鼠随机分为造模组30只、假手术组10只;采用纤维环穿刺法建立椎间盘退变模型,造模组造模成功后随机分为模型组、补肝健腰方组、布洛芬组,每组各8只。造模成功给药6周后检测大鼠髓核组织TNF-α、IL-1β的表达水平。结果模型组腰椎间盘髓核组织TNF-α、IL-1β表达较假手术组增高,差异均有非常显著性意义(P〈0.01);补肝健腰方组腰椎间盘髓核组织TNF-α、IL-1β的含量均低于模型组,差异均有明显统计学意义(P〈0.01);布洛芬组腰椎间盘髓核组织IL-1β的含量低于模型组,差异有统计学意义(P〈0.05)。结论补肝健腰方可能是通过抑制大鼠退变腰椎间盘髓核组织中致炎因子TNF-α、IL-1β的表达来减轻腰椎间盘的退行性改变。
Objective The preventive effects of Bugan Jianyao decoction on TNF-α and IL-1β expressions in rats with lumbar spine degeneration were observed. The efficacy mechanization of the Bugan Jianyao decoction in treatment of lumbar disc degeneration was explored. Methods 40 10-month-old SD rats were randomly divided into model group and the sham-operation group, with 30 and 10 respectively. The lumbar spine degeneration models were established by annulus puncture. After the successful modeling, the rats were randomly divided into model group, Bugan Jianyao decoction group and ibuprofen group, with 8 in each group. The expression levels of TNF-α ,IL-1β in nucleus pulposus of lumbar disc tissue were detected after six-month administration. Results The expression of TNF-α IL-1β in nucleus pulposus of lmnbar disc in the model group was increased compared with sham-operation group, and the differences were very significant (P〈0.01); the expression of TNF-α , IL-1β in nucleus pulposus of the lumbar disc in Bugan Jianyao decoction group were lower than the model group, and the differences were statistically significant (P〈0.01); The level of IL-1β in the ibuprofen group was lower than the model group, and the difference was statistically significant (P〈0.05). Conclusion Bugan Jianyao decoction can reduce degenerative changes in the lumbar intervertebral disc by inhibition TNF-α and IL-1β in nucleus pulposus of the lumbar disc of rats.
