目的 在用酵母双杂交方法将Nelin与人心脏cDNA文库进行杂交筛选,得到3个阳性克隆的基础上,进一步探讨新基因Nelin及其心肌表达产物Nelin蛋白的功能,并寻找能够与其发生相互作用的蛋白质。方法采用基因转染和表达技术,在真核细胞内外分别进行了Nelin蛋白与肌动蛋白(F-actin)结合的共沉降及免疫荧光共定位实验。并通过免疫共沉淀实验进一步验证所得的实验结果。结果 证实了在细胞内外Nelin蛋白均能同F-actin相结合;还能与Filamin的D末端免疫球蛋白样结构发生相互作用。结论 Nelin为一个新的兼有actin及Filamin结合能力的蛋白。在心肌细胞内,Nelin很可能作为一个膜一细胞骨架连接蛋白,参与了细胞黏着斑形成的过程。
Objective To study the function of a novel gene Nelin and its expression protein Nelin in cardiac muscle, and search those Nelin-associated interaction proteins, based on 3 positive cDNA clones acquired by our research group using yeast two hybridization with human adult heart and aorta cDNA libraries. Methods Some bioinformatic softwares were used to ana- lyze the nucleic acid sequence of gene Nelin and the amino-acid sequence of protein Nelin. The co-sedimentation of F-actin with Nelin assay was performed, and immunofluorescence assay was also used to find out the co-localization of Nelin and F-actin. Co-immunoprecipitation was used to further certify the experimental results. Results Nelin protein as a specific cardiac-expressed protein could bind with C-terminus of Filamin besides its binding with F-actin. Conclusion Nelin might play an important role in linking the cytoskeleton to extracellular matrix and participate in the formation of the focal adhesion in cell-matrix junction.
