目的:研究扇贝裙边糖胺聚糖(SS—GAG)抗动脉粥样硬化作用机制。方法:将培养的U937细胞分为正常组、模型组和SS~GAG不同浓度组,除正常组外,其余组加入氧化低密度脂蛋白造模后给予相应浓度药物进行处理,分别采用不同方法测定各组细胞超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH—Px)活性和一氧化氮(NO)含量及Ca^2+水平。结果:各组中SOD含量无显著性差别;SS—GAG组中GSH—Px活性和NO含量与模型组比较明显升高,而细胞内Ca^2+水平则明显降低。结论:SS—GAG可能通过增强GSH-Px活性和NO释放,同时降低细胞内Ca^2+水平而发挥抗动脉粥样硬化作用。
OBJECTIVE: To study the mechanism of anti - atherosclerosis action of glycosaminoglyean from scallop skirt (SS-GAG) . METHODS: The incubated U937 cells were divided into normal group, model group and SS GAG groups (at different concentrations). All groups except the normal group were treated with drugs (at a corresponding concentration) after modeling with oxidized low density lipoprotein (LDL) . The SOD level, GSH-Px activity and NO concentration and the intracellular Ca^2 + in each group were measured respectively by different methods. RESULTS: No significant differences were noted among groups in SOD level. However, as compared with model group, GSH- Px activity and NO concentrations in SS- GAG groups were significantly increased, but the intraeellular Ca^2+ level was significantly decreased. CONCLUSION: The mechanism of the anti - atherosclerosis action of SS- GAG may be attributed to its ability to increase GSH - Px activity and the releasing of NO meanwhile decreasing the intracellular Ca^2 + level in foam cells.
