目的:研究含白花蛇舌草(SpreadingHedvotisHerb,简称SH)血清对植物血凝素(PHA)诱导大鼠脾淋巴细胞增殖、混合淋巴细胞培养(简称MLR)淋巴细胞增殖及对淋巴细胞分泌白细胞介素-2(IL-2)和γ-干扰素(IFN-γ)的影响。方法:大鼠20只,随机分为5组,每组4只,分为SH低剂量(4g/kg)、中剂量(16g/kg)、高剂量(24g/kg)组,含环孢素A(CsA)血清组,按相应药物灌胃,空白血清组用等量生理盐水灌胃。MTT法测定PHA诱导的大鼠脾淋巴细胞和MLR淋巴细胞增殖,ELISA法测定PHA诱导淋巴细胞分泌IL-2、IFN-γ含量。结果:SH高剂量组可抑制PHA诱导大鼠脾淋巴细胞增殖,SH高剂量组、含CsA血清组与空白血清组比较,差异均有显著性意义(P〈0.05)。双向MLR体系中,各剂量组合SH血清均可抑制淋巴细胞增殖,其中SH高剂量组与空白血清组比较,差异有显著性意义(P〈0.05);在单向MLR体系中,各剂量组合SH血清可调节淋巴细胞增殖。SH中剂量组、SH高剂量组血清抑制脾淋巴细胞增殖,与空白血清组比较,差异均有显著性意义(P〈0.05)。含CsA血清组血清可抑制单向、Y,K向MLR体系中淋巴细胞增殖,与空白血清组比较,差异均有显著性意义(P〈0.05)。SH中剂量组、SH高剂量组、含CsA血清组血清均可显著抑制大鼠脾淋巴细胞分泌IL-2、IFN-γ含量,与空白血清组比较,差异均有显著性意义(P〈0.05)。结论:SH能抑制淋巴细胞和混合培养淋巴细胞增殖,其作用机制可能与抑制淋巴细胞分泌IL-2、1FN-γ有关。
Objective= To study the effect of the serum containing Herba Hedyotis Diffusae (HHD) on splenic lymphocytes proliferation induced by phytohemagglutinin (PHA)and in in-vitro mixed lymphocyte reaction (MLR) system as well as the contents of interleukin-2(IL-2) and interferon gamma(IFN-γ). Methods: Twenty rats were randomly divided into five groups, blank group, cyclosporin a(CsA, in the dosage of 15 mg/kg) group, and high(H)-, middle(M)-, Iow(L)-dose HHD groups (in the dosage of 4, 16 and 24g/kg, respectively). The blank group was administrated with normal saline. Lymphocyte proliferation induced by PHA and lymphocyte proliferation in MLR system were detected by means of M-R- assay. The levels of IL-2 and IFN-γ were detected by enzyme linked immunosorbent assay(ELISA). Results: The inhibition of lymphocyte proliferation in H group and CsA group was superior to that in the blank group(P〈0.05). In two-way MLR system, all HHD groups could inhibit the proliferation of lymphocytes, the difference between H group and the blank group being significant (P〈 0.05). In one-way MLR system, all HHD groups could regulate the proliferation of lymphocytes. The inhibition of splenic lymphocytes proliferation in H group and M group was superior to that in the blank group(P〈 0.05). In two-way MLR system, the difference between H group and the blank group was significant(P〈 0.05). In both one-way and two-way MLR system, the inhibition of lymphocyte proliferation in CsA group was superior to that in the blank group(P〈 0.05). The inhibition of IL-2, IFN-γ secretion in H group , M group and CsA group was superior to that in the blank group(P〈0.05). Conclusion: HHB can inhibit the proliferation of lymphocytes and the mixed lymphocytes, and the mechanism is related with the inhibition of IL-2 and IFN-γ by rat iymphocytes
