目的观察谷氨酰胺对高氧肺损伤的干预效果,探讨防治高氧肺损伤的有效措施。方法建立大鼠肺损伤模型,比较高氧组与空气组及不同剂量谷氨酰胺组大鼠肺组织病理形态学的变化以及血浆8-异前列腺素F2 a水平。结果高氧组肺泡细胞形态破坏明显,细胞内结构肿胀,大剂量谷氨酰胺组细胞形态破坏程度较高氧组明显减轻。高氧组各时间点血浆8-异前列腺素F2 a水平均显著高于空气组(P均〈0.05);高氧组血浆8-异前列腺素F2 a水平在3、7、14 d均高于大剂量谷氨酰胺组(P均〈0.05),与小剂量谷氨酰胺组相近(P〉0.05)。结论氧化应激与高氧肺损伤密切相关,大剂量谷氨酰胺对高氧肺损伤的氧化应激作用具有明显的干预作用。
Objective To observe the effect of glutamin on hyperoxia-induced lung injury,and to find a effective method for the protection against hyperoxia-induced lung injury.Methods The models of hyperoxia induced lung injury were established and the lung histopathology was observed by light microscope.The difference of 8-iso-PGF2a in blood plasm between hyperoxia-model group and air group,different dose glutamine groups and hyperoxia-model group was compared.Results In the hyperoxia-model group,the alveolus cells were damaged severely and the structures in alveolus cells were swelling.The damaged degree of cells in hyperoxia + high-dose glutamine group was alleviative than in the hyperoxia-model group.Compared with air group,the level of 8-iso-PGF2a of blood plasm in hyperoxia-model group was higher significantly at every time spot(all P0.05).The level of 8-iso-PGF2a of blood plasm in hyperoxia-model group was higher than that of hyperoxia + high-dose glutamine group on the 3rd,7th and 14th day(all P0.05).Compared hyperoxia-model group with hyperoxia + low-dose glutamine group,the level of 8-iso-PGF2a of blood plasm had no statistically significant at every time spot(all P0.05).Conclusions Oxidative stress reaction is closely related to hyperoxia-induced lung injury.Glutamine has interventional effect on the pathological process of hyperoxia-induced lung injury and its protecting effect has dose dependence.
