【目的】H2O2水溶液可以诱导葡萄叶片中白藜芦醇含量升高,前期研究发现喷洒法诱导效果有限,与叶表面与诱导剂接触不充分有关。研究使用H2O2与表面活性剂SDS、TX-100混合以喷洒的方式诱导葡萄离体叶片,以期改善并提升诱导效果。【方法】以红地球葡萄叶片为试验材料,以表面活性剂SDS、TX-100与H2O2溶液为供试材料。采用H2O2处理离体葡萄叶片,宏观荧光法定性检测白藜芦醇,HPLC法检测白藜芦醇含量的变化,DAB染色法检测氧化胁迫程度。【结果】H2O2溶液处理离体葡萄叶片,可使离体叶片中H2O2含量升高并造成氧化损伤,同时使叶片中白藜芦醇含量升高。表面活性剂SDS、TX-100显著增加了H2O2的诱导作用,也带来了氧化损伤程度加重。以TX-100诱导组含量最高,达到(85.88±0.34)μg/g。【结论】表面活性剂TX-100、SDS对H2O2诱导葡萄叶中白藜芦醇含量有增效作用。
【Objective】H2O2was proved to increase the resveratrol content in grape leaves. But,previous studies found the spray treatment showed a limitation to foliar contamination with the inducer. In order to get better effects on induction,two kinds of surfactants( SDS and TX- 100) were employed with H2O2 in treatments on the detached grape leaves. 【Method】Red globe grape leaves were treated by those surfactants combined with H2O2 aqua solution at different concentrations as the material for testing in the present study. Both qualitative and quantitative assays on resveratrol in the leaves were made by HPLC and fluorescence photographic measurement. The foliar H2O2 content was evaluated by DAB stain. 【Result】The increased foliar resveratrol contents were observed with various H2O2 treatments,so did the level of the oxidative stress in the treatment with the additional SDS or TX- 100 H2O2 foliar treatments,the H2O2 content and the oxidative stress came with the increased resveratrol content significantly. TX- 100 treatment showed the highest induction ratewas(85. 88 ± 0. 34) μg / g.【Conclusion】Both surfactants TX- 100 and SDS showed synergistic effect with H2O2 on the spraying induction of resveratrol in grape leaves.
