目的:探讨累加电针治疗神经源性痛大鼠的作用机制。方法:Wistar大鼠随机分为正常对照组,慢性压迫性损伤(CCI)组,CCI+EA2(天)D组,CCI+EA2(周)w组,去卵巢(OVX)+CCI组,OVX+CCI+EA2D组,OVX+CCI+EA2W组。去卵巢大鼠在手术后每天颈部皮下注射D一半乳糖,连续45天造成神经记忆损伤。结扎坐骨神经造成慢性神经疼痛模型,电针双侧“足三里”一“阳陵泉”穴1次/D,分别电针2D、2W。用RT—PCR的方法检测下丘脑前阿黑皮素(POMC)mRNA和海马、下丘脑孤啡肽前体(PPPNOC)mRNA的表达。结果:与正常对照组比,CCI组下丘脑POMCmR.NA表达水平显著升高(P〈0.05),海马及下丘脑PPNOCmRNA表达水平也显著升高(P〈0.05)。与CCI组比,CCI+EA2W组POMCmRNA表达水平进一步显著上调(P〈0.05),而PPNOCmRNA表达水平显著降低(P〈0.05)。在OVX+CCI动物上,与OVX+CCI组比,OVX+CCI+EA2W组下丘脑POMC的表达量明显上调(P〈0.05),而OVX+CCI+EA2D和OVX+CCI+EA2W海马PPPNOCmRNA的表达显著下调(P〈0.05),但后两组下丘脑PPPNOCmRNA的表达无显著变化(P〉0.05)。结论:电针对慢性神经痛大鼠的累积性镇痛效应与下丘脑POMCmRNA表达的上调和海马PONCmRNA表达的下调密切相关;记忆损伤可减弱电针上调下丘脑POMC基因的表达。
Objective:To investigate the mechanism of repeated electroacupuncture (EA) underlying the cumulative analgesic effect in neuropathie pain rats. Methods : A total of 35 Wistar rats were randomized into control ( n = 5 ) , chronic constrictive injury (CCI) (n=5),CCI+EA2day (D) (n=5),ovaficctomy (OVX) +CCI (n=5),OVX+CCI+EA2D (n=5),OVX+CCI+ EA2 weeks (W) ( n = 5) groups. The OVX rats were subcutaneously injected with D - galaetose (15mg/100g) solution, once daily for 45 days (D). Chronic pain model was established by ligating the left sciatic nerve with pieces of surgical suture. EA(2/ 15Hz 1- 2mA ) was applied to bilateral "Zusanli" ( ST 36) - "Yanglingquan" ( GB 34) for 30 min, once daily for 2D and 2 W, respectively. Hypothalamic Pre - proopiomelanocortin ( POMC ) mRNA expression, and hypothalamic and hippocampal prepronociceptin (PNOC) mRNA expression were detected by reverse transcription polymerase chain reaction ( RT - PCR). Results:In comparison with the normal control group, the hypothalamic POMC mRNA expression was upregulated significantly ( P 〈 0. 05 ) and hypothalamus and hippocampal PPNOC mRNA expression downregulated considerably in CCI group ( P 〈 0. 05 ). Compared with the CCI group, the expression of hypothalamic POMC mRNA was increased markedly in CCI + EA2W group (P 〈 0. 05 ), while the expression of hippocampal PNOC mRNA decreased distinctly in both CCI + EA2D and CCI + EA2W groups (P 〈 0. 05). In OVX + CCI rats, compared with the OVX + CCI group, hypothalamic POMC mRNA expression was upregulated significantly in OVX + CCI + EA2W group ( P 〈 0. 05 ), whereas hippocampal PNOC mRNA expression was downregulated remarkably ( P 〈 0.05).
