为了评价硫酸化修饰后人参皂苷-Rh2的活性变化,利用薄层色谱、红外光谱及质谱鉴定所用Ⅱa、Ⅱb为人参皂苷-Rh2的硫酸化衍生物,并探讨了人参皂苷-Rh_2及其硫酸化衍生物Ⅱa、Ⅱb对小鼠的巨噬细胞吞噬中性红和在LPS刺激下分泌TNF-α量的影响。结果显示,衍生物Ⅱa、Ⅱb在1 mg/L时均能显著促进小鼠腹腔巨噬细胞吞噬活性(P〈0.05),Rh2在0.2mg/L和5mg/L亦可显著促进巨噬细胞吞噬中性红(P〈0.05)。Ⅱa 0.2、5 mg/L时,Ⅱb在为5 mg/L时其在LPS的协同作用下能极显著促进小鼠腹腔巨噬细胞分泌TNF-α(P〈0.01),而Rh_2在5mg/L表现出极显著的抑制LPS促进小鼠腹腔巨噬细胞分泌TNF-α作用(P〈0.01)。结果提示,Rh_2硫酸化修饰后对巨噬细胞的功能有显著增加作用。
In order to evaluate the activities of sulfated derivatives of ginsenoside Rh_2,Ⅱa andⅡb were identified by IR,TLC and Mass spectra.The effects of the derivatives on macrophage competence were determined by phagocytosis of neutral red and TNF-αinduced by LPS in vitro.The data showed thatⅡa andⅡb were the sulfated derivatives of ginsenoside Rh_2.Ⅱa andⅡb at 1 mg/L significantly promoted the phagocytosis of macrophages in mice(P0.05).Rh_2 at 0.2 and 5 mg/L also significantly enhanced macrophage phagocytosis of neutral red(P0.05).Ⅱa at 0.2 and 5 mg/L,andⅡb at 5 mg/L,very significantly promoted the secretion of TNF-αof macrophages induced by LPS(P0.01).But,Rh_2 at 5 mg/L,very significantly inhibited it.The results indicated that the sulfated ginsenosides Rh_2 promoted the macrophage competence.
