弓状核损毁对小鼠ghrelin餐前分泌峰的影响

目的观察损毁新生小白鼠弓状核后是否会引起血浆ghrelin水平的变化。方法取第12周正常对照小鼠和谷氨酸单钠(MSG)处理的小鼠,内眦取血,酶联免疫分析(EIA)法测血浆ghrelin水平;小鼠弓状核冷冻切片,甲酚紫染色观察尼氏体变化;取小鼠胃体黏膜组织,Westen blot检测proghrelin蛋白表达的变化。结果MSG处理组小鼠下丘脑弓状核尼氏体神经元数量明显少于正常对照组小鼠(t=5.21,P〈0.01)。两组小鼠血浆ghrelin水平在禁食前后均有明显变化,但两组小鼠之间比较差异无显著性。与禁食前相比,禁食48 h后两组小鼠胃体黏膜组织中proghrelin蛋白表达水平明显升高,但禁食前后两组小鼠之间比较差异无显著性。结论下丘脑弓状核对MSG处理的小白鼠血浆ghrelin水平的调控并不是必需的。

英文摘要:

Objective To explore whether the plasma ghrelin level would be changed due to destruction of arcuate nucleus in newborn mice. Methods Orbital blood was taken from 12-week-old and monosodium glutamate-treated(MSG) mice.Plasma level of ghrelin was determined with enzyme-linked immune assay(EIA),frozen section of arcuate nucleus with cresyl violet staining used for observing changes of Nissl neurons and Western blot employed to study proghrelin protein expression in the mucosa of mouse gastric body. Results The number of Nissel neuron in arcuate nucleus was significantly reduced in MSG-treated mice compared with that of normal controls(t=5.21,P0.01).There were significant changes of plasma ghrelin level before and after fasting in both groups,but the difference was not statistically different.The level of proghrelin protein expression in gastric mucosa was significantly increased after fasting for 48 h in both groups,but that was not statistically different between the two groups.Conclusion The results suggest that typothalamic arcuate nucleus does not play a critical role in regulating plasma ghrelin levels in MSG-treated mice.
