利用MS和YIM38#培养基、16SRNA、液体发酵法、DNS法和扫描电镜,从“云麻1号”大麻籽中筛选出内生菌,进行菌株分离纯化、菌株鉴定、液体发酵和酶活测试,分析果胶酶溶液处理对木质材料结构的影响。共筛选出7株芽孢杆菌,在pH5.5条件下HS032菌株的果胶酶活性最高,为21.78U/mL利用该果胶酶粗酶液对工业大麻秆进行处理并与4种市购果胶酶的处理效果进行比较,结果表明HS032菌株产生的果胶酶对工业大麻秆中纤维细胞的分离效果最好。果胶酶受到了原料酸碱性的显著影响,在植物内生菌中筛选木材加工专用高效果胶酶是可行的。
The hemp-seed-endophytes were screened, separated, sublimated and identified by MS medium, YIM 38# medium and 16S RNA technology. The pectinase solution was got by liquid fermentation and the pectinase activities were measured by DNS method. Then the pectinase solution from hemp seed endophytes HS032 and other four kind of pectinase bought from market used to treat the hemp stalk and the structure were observed by SEM. The results showed that there were all 7 strains can be used to produce pectinase all were Bacillus and the activities of pectinase produced from strain-HS032 was 21.78 U/mL what was higest under the pH-value was 5.5 and the fiber separation was best after the hemp stalk treated by the pectinase compared with the other four kinds of pectinase, due to that the pectinase activities can be influenced greatly by the pH-value of the raw-materials. Thus, looking for the strains from plant endophytes to produce the pectinase used in wood processing was feasible.
