采用ACQUITY UPLC HSS T3色谱柱,以含0.1%甲酸的乙腈-水为流动相,0.3mL/min梯度洗脱以超高效液相色谱-串联质谱法建立了测定菊花中多酚类化合物的方法。菊花样品经过甲醇水溶液超声提取后直接进样分析,通过质谱分析以及比对标准品或文献中化合物的保留时间和质谱参数等,鉴定了亳菊、滁菊、贡菊和杭菊中10种咖啡酰基奎宁酸类化合物和22种黄酮类化合物。结果表明,4种菊花中多酚类化合物的含量均符合《中国药典》规定的要求,但各种类咖啡酰基奎宁酸和黄酮化合物的组成及含量差异显著,含量较高的除绿原酸(3-CQA),3,5-二咖啡酰基奎宁酸(3,5-DCQA)和木犀草苷(L-7-O-Glu)之外,还有芹菜素-7-O-丙二酰葡萄糖苷(AP-7-O-6义-MGlu)、4,5-二咖啡酰基奎宁酸(4,5-DCQA)、1,5-二咖啡酰基奎宁酸(1,5-DCQA)、木犀草素-7-O-丙二酰葡萄糖苷(L-7-O-6义-MGlu)和芹菜素-7-O-葡萄糖苷(AP-7-O-Glu)等。4种菊花中,黄酮类化合物总量差异较大,但咖啡酰基奎宁酸类化合物总量相当,对多酚类化合物总量而言,贡菊含量最高,达66.85mg/g,其次为杭菊、滁菊和亳菊,含量分别为54.69,44.59和37.10mg/g。
An ultra-high performance liquid chromatography-diode array detection-tandem mass spectrometric (UPLC-DAD-MS/MS) method was developed for determination of phenolic compounds in chrysanthemum samples. The chromatographic separation was performed on an UPLC HSS T3 column by gradient elution with mobile phases of acetonitrile and water both containing 0.1% formic acid at a flow rate of 0.3 mL/min. The samples were extracted with aqueous methanol solution under ultrasonic condition, and the crude extracts were directly assayed without any further pretreatment. Total 10 eaffeyolquinic acids ( CQAs ) and 22 flavonoids were positively identified in Boju, Chuju, Gongju and of samples with standards and publications. The results Hangju by comparing retention times and mass spectra showed that the amounts of phenolic compounds in four chrysanthemum samples all were agreed with the criteria of China Pharmacopoeia (2010), but there were significant differences for the content and constitute distribution of each caffeyolquinic acid and flavonoid. Except chlorogenic acid (3-CQA), 3,5-dicaffeoylquinic acid (3,5-DCQA) and luteolin-7-O-glucoside (L-7- O-Glu) , other compounds with high content, such as apigenin-7-O-6"-malonylglucoside (AP-7-O-6"-MGlu), 4, 5-dicaffeoylquinic acid (4, 5-DCQA), 1, 5-dicaffeoylquinic acid ( 1, 5-DCQA), luteolin-7-O-6"- malonylglucoside (L-7-O-6"-MGlu) and apigenin-7-O-glucoside (AP-7-O-Glu), were also found in chrysanthemum samples. Among the four chrysanthemum samples, there were big variances for the amounts of total flavonoids, but total CQAs were found at the same level; as for total phenolic compounds in four chrysanthemum samples, Gongju showed the highest content of 66.85 rag/g, followed by Hangju, Chuju and Boju with the contents of 54.69, 44.59 and 37.10 mg/g, respectively.
