为了考察6种不同结构黄酮类化合物(5-OH-3′-OCH3黄酮,7-OH-3′-OCH3黄酮,7-OH-2′-OCH3黄酮,3,5,7,4′-4OH黄酮,5,7,3′,4′-4OH黄酮,Neodiosmin)对乳腺癌细胞MCF7的药物敏感性,综合运用MTT法,Hoechst 33342染色法以及流式细胞术评价黄酮类化合物与阿霉素联用对MCF7的抑制效果。结果表明,部分黄酮类化合物细胞毒性较低,其中5-OH-3′-OCH3黄酮,7-OH-3′-OCH3黄酮,3,5,7,4′-4OH黄酮能明显增强阿霉素对MCF7细胞的抑制作用。同时发现黄酮类化合物与阿霉素联用在MFC7细胞上所表现出来的协同作用可能与黄酮类化合物A环上5位与7位的羟基取代相关。
To investigate inhibition effects of Doxorubicine combined with 6different kinds of flavonoids(5-OH-3′-OCH3 flavonoid,7-OH-3′-OCH3 flavonoid,7-OH-2′-OCH3 flavonoid,3,5,7,4′-4OH flavonoid,5,7,3′,4′-4OH flavonoid,Neodiosmin)on MCF7 breast cancer cell,MTT assay,hoechst 33342 dyeing and flow cytometry were applied to assess inhibition effects of Doxorubicine combined with flavonoids.The result shows that some flavonoids have low cytotoxicity,such as 5-OH-3′-OCH3 flavonoid,7-OH-3′-OCH3 flavonoid and 3,5,7,4′-4OH flavonoid which can greatly enhance inhibition effect of Doxorubicine on MCF7 cell.And synergistic effect of Doxorubicine combined with flavonoids on MCF7 cell may be related to hydroxy replacement(5-OH,7-OH)of flavonoids.
