目的探讨地塞米松诱导小鼠胸腺细胞凋亡的适宜剂量。方法小鼠腹腔注射不同剂量(20、40、50mg/kg)地塞米松后12 h处死,提取胸腺细胞DNA,用琼脂糖凝胶电泳方法观察DNA梯形条带(DNA ladder),再用流式细胞术和电镜检测胸腺细胞凋亡情况。结果 40、50 mg/kg地塞米松组出现DNA ladder,流式细胞术发现凋亡峰,电镜观察显示凋亡小体。结论 40 mg/kg地塞米松可诱导小鼠胸腺细胞凋亡。
Objective To study the optimal dose of dexamethasone that induces apoptosis of mouse thymocytes. MethodsThe mice were sacrificed 12 h after intraperitoneal injection with different doses(20, 40 and 50 mg/kg) of dexamethasone. DNA ladder and apoptosis of thymocytes were determined by agarose gel electrophoresis, flow cytometry and electron microscopy, respectively. Results DNA ladder and apoptotic peak and bodies were only present after treatment with 40 mg/kg or 50 mg/kg dexamethasone. Conclusion Dexamethasone of 40 mg/kg can induce apoptosis of mouse thymocytes.
