目的通过化学合成铜绿假单胞菌密度感应(quorum sensing,QS)信号分子OdDHL,并观察该分子对小鼠肥大细胞生物学作用。方法通过化学方法合成OdDHL分子,经质谱、磁共振和高效液相色谱确定其结构和纯度,并采用OdDHI.分子感应菌株检测其生物活性。以不同浓度OdDHL分子作用于小鼠肥大细胞系P815,观察不同时间点细胞活力改变、凋亡现象以及胞内钙离子变化。结果成功合成具有生物活性的OdDHL分子,该分子能以时间、剂量依赖方式抑制细胞活力,并能诱导细胞凋亡,同时可卜调胞内钙离子浓度。结论铜绿假单胞菌OdDHL分子可诱导P815细胞凋亡并上调胞内钙离子浓度。
Objective To investigate the biological effects of Pseudomonas aeruginosa quorum sensing molecule OdDHL on murine mast cells. Methods The molecule structure and purity of synthesized OdDHL were confirmed by mass spectrum or proton nuclear magnetic resonance (NMR) and high-perform- ance liquid chromatography, respectively. Its biological activity was checked using a quorum sensing sensor bacterial strain. The viability, apoptosis and intracellular calcium changes of P815 cell line in response to different concentration of OdDHL were determined. Results The biological active OdDHL was synthesized successfully. OdDHL inhibited proliferation of P815 cells in a dose and time dependent manner. It also induced apoptosis and intracellular calcium release in P815 cells. Conclusion Pseudomonas aeruginosa quorum sensing molecule OdDHL induces apoptosis and intracellular calcium release in murine mast cell line P815.
