目的:观察精制熊胆粉的抗过敏作用,并探讨其可能的作用机制。方法:通过IgE诱导大鼠被动皮肤过敏反应(PCA)实验,用比色法测定熊胆粉在体内对肥大细胞影响。体外实验观察精制熊胆粉对IgE诱导组胺释放、细胞内钙摄入及肿瘤坏死因子-α、白细胞介素6以及NF-κB p65的影响。结果:体内实验显示,精制熊胆粉高(100mg/kg)、中(50mg/kg)剂量组能明显抑制大鼠PCA。体外实验显示,高(100μg/L)、中(50μg/L)浓度精制熊胆粉明显抑制了肥大细胞脱颗粒、组胺的释放、细胞内钙摄入以及肿瘤坏死因子-α、白细胞介素6以及NF-κB p65蛋白表达。结论:精制熊胆粉可能通过抑制NF-κB阻止肥大细胞的活化。
Obiective:To investigate the anti-allergic effects of the refined bear gall(RBG) and to explore its possible mechanism.Methods:The experiment of passive cutaneous anaphylaxis was used to determine the effect of RBG on degranulation of mast cells in vivo.For in vitro study,various doses of RBG were added to of sensitized rat peritoneal mast cells(RPMC).The IgE-induced release of degranulation was measured by enzymatic assay,histamine by EIA and cytokines by ELISA.Results:Treatment with RBG(100mg/kg)and moderate dose of Anthocyanidin(50mg/kg)was followed by a decrease in PCA of rats.whereas results in high dose group(100μg/L)and moderate dose of RBG group(50μg/L) indicated significantly inhibitory effect on histamine release,the calcium uptake、TNF-α、IL-6、NF-κB p65 of rat peritoneal mast cells(RPMC) induced by antigen(P0.05).Conclusion:RBG night suppress the anaphylaetic reaction through inhibition of NF-κB p65.
