目的观察创伤后应激障碍(PTSD)模型大鼠海马神经元分子伴侣葡萄糖调节蛋白94(GRP94)的表达变化。方法建立单一延长应激(SPS)大鼠模型,随机分为SPS刺激后第7天组、第14天组及正常对照组,采用免疫组化、Western blot、RT-PCR方法检测各组海马神经元GRP94的表达。结果免疫组化及Western blot结果均显示:造模第7天海马神经元GRP94蛋白表达较正常对照组明显增加(P〈0.05),而第14天明显降低(P〈0.05)。RT-PCR结果显示:造模第7天海马神经元GRP94 mRNA水平明显增加(P〈0.05),第14天则明显降低。结论 PTSD大鼠海马GRP94蛋白水平发生了变化,为进一步研究内质网径路细胞凋亡提供了实验基础。
Objective To observe the changes of molecular chaperone glucose-regulated protein 94 (GRP94) in the hippocampus of post- traumatic stress disorder rat model. Methods Totally 45 rats were randomly assigned to three groups including a normal control group(nd, n =15) and two SPS groups (sps-7d,n =lS;sps-14d,n =15),and prepared for single-prolonged stress (SIX3) model. Immunohistochem- istry,Westem blot and reverse transcfiption-polymerase chain reaction (RT-PCR) were performed to examine the changes of GRP94 in the rat hippocampus. Results By using immunohistoehemistry and Western blot, the expression of GRP94 in the group of sps-7d was markably increased compared with the normal control group (P 〈 0.05 ). The expression of GRP94 in sps-ldd group showed a significantly decreased compared to the sps-7d group (P 〈 0.05). The mRNA expression of GRPgd in RT-PCR had the similar trends of changes. Conclusion SPS would induce changes of GRP94 expression in the rat hippocampus, which provided basis for future study of neuronal apoptosis induced by endoplasmic reficulum stress.
