目的:观察黄芪皂甙和丹参酮ⅡA注射液对大鼠骨骼肌钝挫伤后修复过程中氧化应激的影响。方法:120只雄性SD大鼠随机分为黄芪皂甙组、丹参酮ⅡA注射液组、黄芪皂甙+丹参酮ⅡA注射液组、黄芪丹参注射液组、生理盐水对照组,每组24只。采用打击装置造成右侧腓肠肌中段钝挫伤模型,分别于损伤局部注射黄芪皂甙、丹参酮ⅡA注射液、黄芪皂甙+丹参酮ⅡA注射液(黄芪皂甙0.5 ml+丹参酮ⅡA注射液0.5 ml混合)、黄芪丹参注射液(黄芪注射液0.5 ml+丹参注射液0.5 ml混合)及生理盐水,注射剂量均为1 ml/只/次,以后每隔3天注射一次,并于损伤后1、4、7和14天取腓肠肌测定丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性、谷胱甘肽过氧化物酶(GSH-px)活性。结果:与生理盐水对照组相比,丹参酮ⅡA注射液组、黄芪皂甙+丹参酮ⅡA注射液组、黄芪丹参注射液组MDA含量在第4天时显著降低(P〈0.05),而黄芪皂甙组下降不显著;与生理盐水对照组相比,黄芪皂甙组、丹参酮ⅡA注射液组、黄芪皂甙+丹参酮ⅡA注射液组、黄芪丹参注射液组SOD、GSH-px活性在第4天时显著升高(P〈0.05),黄芪皂甙组升高幅度低于丹参酮ⅡA注射液组、黄芪皂甙+丹参酮ⅡA注射液组、黄芪丹参注射液组,与黄芪丹参注射液组相比,丹参酮ⅡA注射液组、黄芪皂甙+丹参酮ⅡA注射液组SOD、GSH-px活性无显著差异(P〉0.05)。结论:黄芪皂甙和丹参酮ⅡA注射液均可减轻骨骼肌钝挫伤后氧化应激的损伤,并提高骨骼肌SOD和GSH-px的活性,而丹参酮ⅡA注射液作用更为显著。
Objective To investigate the effects of Chinese medicine Astragalosides and TanshinoneⅡA on oxidative stress in rat skeletal muscles after acute contusion.Methods One hundred and twenty male Sprague-Dawley rats were randomly divided into five groups:Astragalosides group;Tanshinone IIA group;Astragalosides and Tanshinone IIA group;HuangqiDanshen group;physiological saline group.The acute skeletal muscle contusion model was produced at the right gastrocnemius by specific device.Local injection at the injury site with Astragalosides 1ml in Astragalosides group;Tanshinone IIA 1ml in Tanshinone IIA group;Astragalosides and Tanshinone IIA(A mixture of Astragalosides and Tanshinone IIA with each 0.5 ml) in Astragalosides and Tanshinone IIA group;HuangqiDanshen(A mixture of Huangqi injection and Danshen injection with each 0.5 ml)in HuangqiDanshen group;physiological saline 1ml in physiological saline group.The injection dosage was 1ml per capita per time.The injection continued every three days until sample collection.The muscles were sampled at the day 1,4,7 and 14 after injury respectively,and then investigated the parameters of oxidative stress,namely MDA and the activities of antioxidant enzymes(SOD and GSH-px) in gastrocnemius after contusion.Results Compared with the physiological saline group,The concentrations of MDA in Tanshinone IIA group,Astragalosides and Tanshinone IIA group,HuangqiDanshen group were significantly lower at 4 days after injury(P 0.05).But The concentrations of MDA in Astragalosides group had no change(P 0.05).Compared with the physiological saline group,the SOD and GSH-px activities in Astragalosides group,Tanshinone IIA group,Astragalosides and Tanshinone IIA group,HuangqiDanshen group were significantly increased at 4 days after contusion(P 0.05).However an elevation of SOD and GSH-px activities in Astragalosides group were lower than Tanshinone IIA group,Astragalosides and Tanshinone IIA group,HuangqiDanshen group.Compared with HuangqiDanshen group,the SOD and
