基于分子吸收和发射测量,研究了盐酸小檗碱与特定序列DNA间的相互作用。结果发现双链DNA中含有不配对G碱基时,其与盐酸小檗碱作用后产生的光谱特征将明显不同于完全互补的双链DNA。该特征可被用于检测1个错配碱基的双链DNA。利用Hyperchem Professional软件包计算了4种寡聚核苷酸的电荷分布情况,并在此基础上进一步探讨了盐酸小檗碱与寡聚核苷酸之间的作用机理。
The interaction of berberine hydrochloride (BBH) with sequence-specific DNA was studied by measuring their molecular absorption and fluorescence spectra. It was found that the characteristic spectrum produced by the interaction of BBH with double-bond DNA containing the unpaired G bases was significantly different from that with the complementary double-bond DNA. The characteristic spectrum could be used to detect the double-bond DNA with one base mispairing. Based on the charge distribution of four kinds of oligonucleotides calculated with the software package of Hyperchem Professional, the mechanism of the interaction of BBH with oligonucleotides was further revealed.
