TNFAIP1是第一个被鉴定能受TNF-α(tumor necrosis factor-α)诱导的蛋白.TNFAIP1基因是一个多功能基因,参与DNA的复制和修复,细胞周期的调控,细胞信号通路转导的调节等.采用酵母双杂交的方法以TN-FAIP1为靶蛋白筛选得到与其相互作用蛋白RIN3.RIN3含有RIN家族多个多功能的结构域,根据RIN3结构域对其分段,研究与TNFAIP1的相互作用.通过免疫共沉淀,荧光共定位一系列细胞证明TNFAIP1蛋白能直接与RIN3相互作用,提示TNFAIP1可能参与细胞的胞吞胞吐.
TNFAIP1 protein is the first identified TNF-α inducible protein.TNFAIP1 is a multifunctional gene involved in DNA replication,DNA repair,cell cycle and cell signal pathway.In this paper,RIN3(Ras interference 3),that contain many functional domains shared with other RIN members and additional Pro-rich domains,was identified in a two-hybrid screen of a human fetal brain cDNA library by using TNFAIP1 as bait.Mutations of RIN3 were cloned,and then the interactions with TNFAIP1 were investigated.Co-immunoprecipitation and Subcellular localization studies demonstrated that TNFAIP1 interacts directly with RIN3,suggesting that TNFAIP1 may participate in cell phagocytosis and exocytosis.
