目的探讨熊果酸对胶原诱导性关节炎(CIA)模型大鼠的抗炎及骨保护作用。方法建立SD大鼠CIA模型,造模成功后随机分为正常对照组、CIA模型组、熊果酸组10 mg/(kg.d)、甲氨蝶呤组0.9 mg/(kg.w)、地塞米松组1 mg/(kg.3 d),分别给药。每周观察各组的关节炎指数和足爪肿胀度,于给药35 d后对大鼠关节X线摄片,后处死大鼠,取足关节制作切片行HE染色并观察病理改变。结果与CIA模型组比较,各治疗组大鼠关节炎指数显著降低;熊果酸组大鼠关节病理指数及关节X线评分较CIA模型组明显降低(P〈0.01),效果与甲氨蝶呤组、地塞米松组相当(P〉0.05)。结论熊果酸能明显缓解CIA大鼠症状、控制炎症,减轻关节的病理改变及骨质破坏。
Objective To investigate the anti-inflammatory and bone protective effects of ursolic acid(UA) on collagen-induced arthritis(CIA) rats.Methods CIA SD rat model was established and these modeling rats were randomly divided into control group,model group,ursolic acid group 10 mg/(kg.d),methotrexate group 0.9 mg/(kg.w) and dexamethasone group 1 mg/(kg.3 d).The rats were treated with corresponding medications.The arthritis index and paw swelling of rats were observed and recorded weekly.The joints of these rats were assessed by X-ray,and then they were sacrificed and the paws were taken for biopsy after administration for 35 days.Results Comparedwith the model group,the arthritis indices of the groups treated with medications were significantly reduced,pathological indices and joint X-ray scores of ursolic acid group were significantly reduced(P〈0.01) as the same effects as methotrexate and dexamethasone.Conclusion UA can significantly alleviate the symptoms of CIA rats,control inflammation and reduce joint pathological changes and bone destruction.
