目的观察单宁酸(tannic acid,TA)对糖尿病(DM)大鼠抗氧化能力及体外蛋白质非酶糖基化作用的影响。方法 70只雄性Wistar大鼠随机分为正常对照组、模型组、氨基胍(AG)组和TA低高剂量组,给药10周后处死大鼠。化学比色法检测各组大鼠血清及肾皮质MDA含量及GSH-Px、SOD、CAT活性。将单宁酸加入到体外建立的非酶糖基化体系,孵育2个月,荧光法进行AGEs测定并计算蛋白质糖基化抑制率。结果单宁酸可降低DM大鼠血清及肾皮质MDA含量,提高GSH-Px、SOD、CAT活性并呈浓度依赖性抑制体外蛋白质非酶糖基化反应。结论单宁酸可提升糖尿病大鼠的抗氧化能力,并具有抑制体外蛋白质非酶糖基化的作用。
Objective To investigate the effect of tannic acid on antioxidant capacity in diabetic rats and non-enzymatic glycation of protein in vitro.Methods Seventy male wistar rats were randomly divided into 5 groups:normal control group,diabetic model group,aminoguanidine(AG) group and low and high dosages of TA groups.All animals were sacrificed at the 10th week after drugs injection.The content of MDA and the activities of GSH-Px,SOD and CAT in serum and renal tissue were measured.Tannic acid was added to system of non-enzymatic glycation.After 2 months of incubation,AGEs were measured by fluorescence method and the inhibition ratio of non-enzymatic glycation of protein was calculated.Results The MDA content was lowered and the the activities of GSH-Px,SOD and CAT were improved by tannic acid in serum and renal tissue of diabetic rats.In vitro,tannic acid inhibited non-enzymatic glycosylation of protein in a concentration dependent.Conclusion These results suggest that tannic acid can increase antioxidant capacity of diabetic rats and inhibit non-enzymatic glycosylation of protein.
