Vav1蛋白是特异于造血细胞表达的鸟苷酸转换因子,它参与了多种免疫细胞如T细胞,B细胞,巨噬细胞,自然杀伤性细胞等的胞内信号转导,发挥着重要作用.小干扰RNA(small interfering RNA,siRNA)是近年来广泛使用的一种降低/消除细胞内蛋白表达的技术.基于这一技术,建立了Vav1干扰RNA载体系统,此系统能够有效的降低白血病细胞系Jurkat细胞内Vav1蛋白的表达,并且使用方便成本低廉.这一系统的成功建立使在多种免疫细胞系中对Vav1的功能进行比较研究成为了可能.
Vav1 protein is a guanine nucleotide exchange factor which specifically expressed in hematopoietic cells.Vav1 has been reported to beinvowed in signal transduction in many kinds of immune cells including T cells,B cells,macrophages and nature killer cells,and plays important roles.Recently,as a new technology used to knockdown or silence protein expressionin cells,small interfering RNA(siRNA) technology has been rapidly developed and extensively applied in protein function research.Based on small interfering RNA technology, we constructed a Vav1 small interfering RNA expression system which can continuously express Vav1 siRNA in mammalian cell lines and efficiently silence Vav1 expression.Successfully construction of this Vav1 small interfering RNA expression system enables us to comparatively investigate Vav1's function in different immune cell lines.
