白术提取物对大鼠预防性调血脂及保肝作用的实验研究

目的:研究白术总提取物和二个不同部位(100%乙醇和50%乙醇部位)对大鼠调血脂和保肝的作用,并确定白术的有效部位。方法:采用高脂模型-预防性给受试物法,给予大鼠白术3种不同提取流份灌胃4周,测定体重和血清中胆固醇(TC)、甘油三酯(TG)和高密度脂蛋白(HDL-C)的含量以及谷丙转氨酶(ALT)和谷草转氨酶fAST)的活性,并观察肝脏病理切片。结果:白术总提取物和100%乙醇部位能显著降低小鼠体重值和TC、TG、动脉硬化指数(AI)水平(P〈0.01)。白术总提取物和100%乙醇部位都具有升高高脂血症大鼠血清中HDL-C水平(P〈0.05或P〈0.01)的作用,但只有100%乙醇部位降低LDL-C(P〈0.01)。白术总提取物,100%乙醇和50%乙醇部位都能拮抗ALT、AST活性的升高(P〈0.01),并且白术总提物组和100%乙醇部位组高脂血症大鼠的肝系数得到了显著性改善(P〈0.05)。结论:白术中亲脂性天然产物(100%乙醇部位)具有较强的降血脂作用,白术总提取物和亲脂性天然产物(100%乙醇部位)都有一定的保肝作用。

英文摘要:

Objective. To investigate preventive effects of Atractylodes macrocephala Koidz Decoction (AKD)and its two fractions (100% alcohol and 50% aqueous alcohol) on the levels of plasma lipid and liverin rat, and define the. bioactive fraction of AKD. Methods: The experiment used the model of metabolic disturbance of lipid-preventive tested sample. The rats were divided into six groups: control, high fat diet (HFD), positive HFD control, water extract from AKD treatment, 100% ethanol fraction treatment and 50% ethanol fraction treatment. Serum levels of TC, TG and HDL-C were measured. Also, activities of ALT and AST were determined. Results: Compared with HFD control group, AKD and itsl00% ethanol fraction significantly reduced TC, TG, AST, ALT and the arteriosclerosis Index (AI) (P〈0.01). AKD and its 100% fraction increased the levels of serum HDL-C (P〈0. 05 or P〈0. 01), but only 100% fraction reduced level of serum LDL-C (P〈0. 01). All AKD and its two fractions could suppressed the increase of ALT, ASTs activities (P~0. 05), and AKD and its 100% fraction improved the ratio of liver and body weigh (P〈0.05). Conclusion: Non-polar natural products (100% ethanol fraction) from AKD showed significant effect on the regulation of serum lipid levels, and both AKD and its non-polar.natural products (100% ethanol fraction) protected the liver in hyperlipidemic rats.
