为了高效利用桑叶资源获取功能活性物质,在连续提取桑叶多糖、黄酮和生物碱的基础上,对3类活性物质进行分离纯化,研究大孔树脂和阳离子交换树脂对桑叶活性物质的吸附与洗脱性能。从3种大孔树脂中筛选的D101大孔树脂对桑叶多糖和黄酮具有较好的分离纯化效果:对桑叶多糖的最大吸附量达到308.42 mg/g,解吸率达70.18%,对桑叶多糖的脱色率和脱蛋白率分别达到89.29%、78.74%;对桑叶黄酮的最大吸附量达到38.11 mg/g,解吸率达81.46%。从2种阳离子交换树脂中筛选001*7阳离子交换树脂分离纯化桑叶生物碱,其最大吸附量达26.61 mg/g,解吸率达83.49%。分离纯化得到的桑叶多糖、黄酮和生物碱样品纯度分别为35.73%、38.64%、29.75%,表明联合使用D101大孔树脂和001*7阳离子交换树脂能有效地从桑叶提取物中分离纯化桑叶多糖、黄酮和生物碱3类活性物质。
In order to obtain active functional substances from mulberry leaf resource efficiently,based on continuous extraction of polysaccharide,flavone and alkaloid from mulberry leaves,we separated and purified these three active substances to study the adsorptivity and elution property of macroporous resin and cation exchange resin to mulberry leaf active substances.The results showed that,among three kinds of macroporous resin for test,D101 had the best separation and purification effect to polysaccharide and flavone from mulberry leaves.The maximum adsorption capacity of D101 to mulberry leaf polysaccharide reached 308.42 mg/g and desorption rate was 70.18%.The rate of decolorization and deproteinization of D101 to mulberry leaf polysaccharide was 89.29% and 78.74%,respectively.The maximum adsorption capacity of D101 to mulberry leaf flavone reached 38.11 mg/g and the desorption rate was 81.46%.Among two kinds of cation exchange resin for test,001 *7 had better separation and purification effect to mulberry leaf alkaloid.The maximum adsorption capacity of 001 *7 to mulberry leaf alkaloid reached 26.61 mg/g and the desorption rate was 83.49%.The purity of polysaccharide,flavone and alkaloid separated and purified from mulberry leaves reached 35.73%,38.64% and 29.75%,respectively.These results indicated that polysaccharide,flavone and alkaloid could be separated and purified from mulberry leaf extract efficiently with macroporous resin D101 combined with cation exchange resin 001 *7.
