目的 应用酵母双杂交技术筛选获得与NF--κB p50亚基同源结构域(RHD)相互作用的多肽。方法 应用酵母双杂交技术,以NF-κB p50 RHD为诱饵,筛选16肽cDNA文库,寻找与其相互作用的多肽,应用β-GAL实验、酵母交配实验及一对一酵母回复性杂交等方法筛除假阳性克隆,确定阳性克隆。结果经酵母双杂交及反复的排查,共获得8个阳性克隆。通过GenBank进行搜寻。未发现与之相匹配的蛋白序列。结论 获得8个与NF-κB p50 RHD相互作用的新型多肽cDNA,多肽的功能需进一步验证。
Objective To obtain polypeptides interacting with NF-κB p50 Rel homology domain(RHD) by yeast two-hybrid technique.Methods Using NF-κB p50 RHD as bait, a 16-peptide cDNA library was screened and the polypeptides interacting with NF-κB p50 RHD were searched.The false positive clones were discarded by β-GAL assay, yeast mating, and one to one yeast two- hybrid methods. Results 8 truly positive clones were obtained. The gene sequences of 8 polypeptides were different with each other and their homologous proteins were not found from GenBank.Conclusion 8 new polypeptides cDNAs have been obtained and their functions need to be validated in the future.
