研究咪达唑仑对山羊不同脑区cAMP含量的影响,探讨其在中枢麻醉中的作用机制。试验用15只健康山羊,3只为生理盐水对照组,其余为试验组,试验组山羊肌肉注射14mg/kg体重咪达唑仑。分别在给药后诱导期、麻醉期、恢复1期和恢复2期4个时间点,每个给药点剖杀3只山羊取脑组织,并采用ELISA测定不同脑区cAMP含量。注射咪达唑仑后,山羊大脑皮层、海马、丘脑、小脑和脑干内cAMP含量显著升高,恢复2期上述脑区内cAMP含量均恢复到正常水平,这种变化趋势与山羊肌肉注射咪达唑仑麻醉后行为学变化基本一致。表明咪达唑仑的麻醉作用可能与激活山羊各个脑区内cAMP信号转导系统相关。
To study the effect of midazolam on c AMP content in different encephalic regions of goats and to explore its central mechanism,we used 15 healthy goats with three of them as saline control group and the rest as experimental group.The goats in experimental group were intramuscularly injected with 14 mg/kg midazolam.Goats in experimental group were executed respectively in induction period,anesthesia period,recovery-form-anesthesia period I and recovery-form-anesthesia period II after induction and the brain tissues were taken immediately.CAMP content was measured by ELISA in different brain areas.The results showed that the c AMP content in cerebrum,hippocamp,thalamus,cerebellum and brainstem was significantly increased and was restored to normal levels in recovery-form-anesthesia period I and recovery-form-anesthesia period II.This trend was consistent with behavioral change of goats after midazolam anesthesia.The anesthesia effect of midazolam may be related to activating the c AMP signal transduction system in various encephalic regions of goats.
