目的:检测食管鳞状细胞癌(ESCC)组织中锌α2糖蛋白(AZGP1)及mRNA的表达。方法:采用RT-PCR检测55例ESCC组织及癌旁正常食管黏膜组织中AZGP1mRNA的表达,免疫组织化学方法检测87例ESCC及54例癌旁正常食管黏膜组织芯片中AZGP1蛋白的表达。结果:ESCC组织中AZGP1mRNA的阳性表达率(30.9%)低于癌旁正常食管黏膜组织(90.9%,χ2=27.676,P〈0.001);AZGP1mRNA的阳性表达与ESCC的部位(P=0.144)、分化程度(P=0.025)及TNM分期(P=0.011)有关。ESCC组织中AZGP1蛋白的阳性表达率(33.3%)明显低于癌旁正常食管黏膜组织(98.1%,χ2=57.519,P〈0.001)。结论:AZGP1mRNA和蛋白的表达降低与ESCC的发生发展有关,AZGP1可能为ESCC的抑癌基因。
Aim:To investigate the expression of zinc-α2-glycoprotein(AZGP1) in esophageal squamous cell carcinoma(ESCC) tissue.Methods:The expression of AZGP1 mRNA in 55 cases of ESCC and the matched normal mucous tissue was detected by reverse transcription PCR.The expression of AZGP1 protein in tissue microarray of 87 cases of ESCC tissue and 54 cases of normal mucous tissue was detected by immunohistochemistry.Results: The positive rate of AZGP1 mRNA expression in ESCC tissue was significantly lower than that in the matched normal tissue (30.9% vs 90.9%,χ2=27.676,P0.001);the positive expression of AZGP1 mRNA was associated with tumor location(P=0.144),cell differentiation (P=0.025),and TNM stage(P=0.011) in ESCC tissue.The positive rate of AZGP1 protein expression in ESCC tissue was significantly lower than that in normal tissue(33.3% vs 98.1%,χ2=57.519,P0.001).Conclusion: The low expression of AZGP1 mRNA and protein may be involved in the carcinogenesis and development of ESCC.AZGP1 gene may be a novel candidate tumor suppressor gene for ESCC.
