目的研究芍药苷(Paeoniflorin,Pae)对重组人白细胞介素1α(rhIL-1α)诱导的人成纤维样滑膜细胞(FLS)功能的影响。方法采用组织块培养法分离培养正常人FLS,四甲基偶氮唑蓝(MTT)法检测FLS的增殖能力,放射免疫法(RIA)检测FLS产生的TNF—α含量,酶联免疫吸附法(ELISA)检测FLS产生的PGE2和cAMP水平。结果在rhIL-1α(0.01、0.1、1、10、100μg·L^-1)的刺激下FLS增殖能力增强,产生的TNF-α和PGE,水平均不同程度升高,cAMP水平下降。Pae(10^-8、10^-7、10^-6、10^-5、10^-4mol·L^-1)体外作用可不同程度抑制由rhIL-1α诱导的FLS增殖,降低FLS产生的TNF—α和PGE2水平,升高cAMP水平。结论rhIL-1α体外刺激可以促进正常人FLS的增殖和分泌功能;Pae可以逆转rhIL-1α对人FLS功能的影响。
Aim To investigate the effects of Paeoniflorin (Pae) on human fibroblast-like synoviocytes (FLS) under the stimulation of rhIL-1α. Methods The joint FLS were isolated from the healthy human and cultured as tissue culture method. The proliferation of FLS was assessed by MTT assay. Level of TNF- α was examined by radio immunoassay (RIA), and levels of PGE2 and cAMP were determined by ELISA. Results The proliferation of FLS was stimulated by different concentrations of rhIL-1α ( 0. 01, 0. 1, 1, 10,100 μg· L^-1) , levels of TNF-α and PGE2 were increased, whereas level of cAMP was decreased. Pae (10^-8, 10^-7, 10^-6, 10^-5, 10^-4 mol· L^-1) treatment inhibited the proliferation of FLS, down-regulation of TNF-α and PGE2 levels as well as up-regulation of cAMP level under the stimulation of rhIL-1α. Conclusion Pae can reverse the proliferation and secretion function of the human FLS induced by rhIL-1α in vitro.
