以9个大白菜品种的花蕾为试材,研究了基因型、6-BA以及不同固体培养基对大白菜小孢子胚发生的影响,以期为其它十字花科蔬菜小孢子培养体系的建立提供技术参考。结果表明:不同基因型大白菜的小孢子胚诱导率具有明显差异,9个大白菜品种中有6个诱导出胚状体,‘北京特好吃火锅菜’出胚率最高,达到每蕾12.38胚;添加0.3mg/L的6-BA可以促进小孢子胚状体的形成,以‘北京特好吃火锅菜’提升效果最显著;添加0.01g/i。活性炭的MS固体培养基比135培养基更适于成熟胚状体发育成苗。
Taking flower buds of 9 Brassica rapa ssp. pekinensis eultivars as materials,the effect of genotypes,6-BA and different solid medium on microspore embryogenesis were studied. The results showed that the embryogenesis rate was significantly different among cultivars. Six cultivars were induced to form embryoids and the highest yield was produced in 'Beijing lhhaochihuoguocai ' with 12. 38 embryoids per bud. Medium supplemented with 0. 3 mg/L 6-BA could promote the embryogenesis rate,especially for 'Beijing Tehaoehihuoguoeai '. The solid MS medium supplemented with 0. 01 g/L active carbon was better than B5 solid medium for plantlet formation from embryoids.
