目的：观察右归丸、淫羊藿、淫羊藿苷含药血清对成骨细胞增殖及Wnt／β-catenin信号通路的作用。方法：获取新生SD乳鼠的成骨细胞，体外培养至第3代细胞，分对照组、右归丸组、淫羊藿组、淫羊藿苷组共4组，分别在含药血清培养后1、3、5、7、9d时用WST法测细胞增殖情况；培养48h后，以In-eellwesternblot的方法检测各组成骨细胞wnt3a、wnt7b、β-catenin蛋白的表达情况。结果：与对照组比较各组均能促进成骨细胞的增殖，右归丸组在5d、7d、9d时差异有统计学意义，淫羊藿组在7d、9d时差异有统计学意义，淫羊藿苷组在9d时差异有统计学意义（P〈0．05）。与对照组比较，右归丸组能显著提高成骨细胞Wnt3a、Wnt7b、pcatenin蛋白表达，差异有统计学意义（P〈0．05）。淫羊藿组及淫羊藿苷组虽能提高Wnt7b的蛋白表达，但与对照组比较，差异无统计学意义（P〉0．05）。同时，淫羊藿组、淫羊藿苷与右归丸组比较，在对Wnt3a、wnt7b、β-catenin蛋白表达的作用上比较差异均有统计学意义（P〈0．05）。结论：右归丸、淫羊藿、淫羊藿苷三者均能促进体外培养的大鼠成骨细胞的增殖，以右归丸作用最显著；右归丸可显著促进wnt3a、wnt7b、β-catenin蛋白的表达。

Objective：To investigate the effects of medicated serum of Yougui pill, epimedium and icariin on the proliferation and expression of related proteins of Wnt/beta-catenin signal pathway of osteoblast. Methods： Osteoblasts were obtained from the new born SD rats and cultured in vitro. The cells of third passage were used. They were divided into 4 groups： control group, Yougui pill group, epimedium group and icariin group. WST assay was adopted to observe the influence to the proliferation of osteoblast after incubation of 1, 3, 5, 7 and 9 days. In-cell western blot assay was used to observe the influence of each group on the expression of wnt3a, wnt7band β-catenin of osteoblast after incubation of 48h. Results： The proliferation of osteoblast was increased in each experiment group compared with the control group. There was significantdifference in Yougui pill groups at 5, 7, and 9 days, in epimedium group at 7 and 9 days, and in icariin group at 9 days （P〈0. 05）. The expression of wnt3a, wnt7b and β- catenin of osteoblast was significantly increased in Yougui pill group compared with control group （P~0.05）. The expres- sion of wnt7b of osteoblast was increased in both epimedium group and Icariin group, but there was no significant differ- enee compared with the control group （P〈0.05）. Compared with Yougui pill group, there was signifieant difference in ex- pression of wnt3a, wnt7b, and β-eatenin of osteoblast in epi- medium group and Icariin group （P〈0. 05）. Conclusion： Yougui pill, epimedium and icariin can significantly increase the proliferation of osteoblast, among which Yougui pill has the best effect. Yougui pill can significantly increase the ex- pression of wnt3a, wntTb, and β-catenin of osteoblast.
