目的:研究针刺对2型糖尿病大鼠胰岛β细胞形态结构的作用。方法:将造模成功的大鼠按随机化原则分为针刺组、西药组、安慰剂组,同时将普食组大鼠作为正常对照组,每组10只。针刺组大鼠取"后三里"(相当于足三里)、内庭和胰俞穴给与针刺治疗,西药组用罗格列酮(0.2mg/kg体质量)灌胃,安慰剂组用双蒸水(2mL/kg体质量)灌胃,1次/d,连续4周。观察治疗前后空腹血糖、血脂变化,实验结束时,分离胰腺,石蜡包埋、酒精脱水及胰岛素(Ins)免疫组化染色后,光镜观察胰岛β细胞形态及胰岛细胞胰岛素表达。结果:针刺组胰腺腺泡、胰岛结构完整,胰岛内细胞排列大体规整,大小、形态接近正常,胞浆肿胀、纤维组织增生少见,改变显著优于西药组而接近正常组;胰岛细胞胰岛素表达针刺组显著高于安慰剂组。结论:针刺具有保护胰岛β细胞形态的作用。
Objective: To study the effect of acupuncture morphologic changes of islet β-cell in type 2 diabetes rats. Methods: Model rats were divided randomly into acupuncture group (n=10), rosiglitazone control group (n=10) and placebo group (n=10). 10 male normal rats were used as normal control group. The acupuncture group rats were selected points of ‘Housanli', ‘Neiting' and ‘Yishu' to treatment with needling. The rosiglitazone group rats were given rosiglitazone (0.2mg/kg) intragastrically, the placebo group rats were given double distilled water (0.2mg/kg) intragastrically, once daily, for 4 consecutive weeks. The change of fasting blood glucose was determined before and after the treatment. Morphological change of pancrease and expression of insulin in islet β -cell were observed with light microscop. Results: Complete structure of pancreatic acinus and islet, arrangement structured of islet ceils, cells size and shape were close to normal, swelling of bubble pulp and fibroustissue hyperplasia were less see on acupuncture group. Changes of acupuncture group were significantly well than that of rosiglitazone group and close to normal control group. Insulin expression of islet cells of acupuncture group were significantly high than that of placebo group. Conclusion: Acupuncture has protective effects of morphology of islet β-cell in type 2 diabetes rats.
