目的:探讨生血增白汤对骨髓移植后小鼠骨髓中干细胞因子(stemcellfactor,SCF)表达水平的影响及其促进骨髓造血重建的作用机制。方法:建立小鼠骨髓移植模型。将实验动物随机分为5组:正常组(未做任何处理)、模型对照组、G-CSF组、生血组和G-CSF+生血组。其中,生血组及G-CSF+生血组胃饲生血增白汤浓缩煎剂。骨髓移植后的第9、14和24天,采用免疫组织化学法检测SCF表达水平,计数外周血细胞和骨髓有核细胞,并进行骨髓组织学观察。结果:骨髓移植后第14天时,生血组外周血细胞中血红蛋白水平高于模型对照组及G-CSF组(P(0.05),G-CSF+生血组血红蛋白水平高于模型对照组(P(0.05);骨髓移植后第24天时,生血组外周血细胞中白细胞水平高于模型对照组(P(0.05)。骨髓移植后第24天时,G-CSF组、生血组和G-CSF+生血组的骨髓有核细胞计数均高于模型对照组(P(0.01)。骨髓移植后第9天时,生血组骨髓造血组织容量及SCF的表达水平均高于模型对照组(P(0.01)。结论:生血增白汤能促进骨髓移植后骨髓中SCF的表达,这可能是生血增白汤重建造血的机制之一。
Objective:To investigate the effect of Shengxuezengbai Decoction on the expression of stem cell factor(SCF)in bone marrow tissues and on the mechanism of hematopoietic reconstitution after bone marrow transplantation(BMT).Methods:The experimental animals were randomly divided into 5 groups:the normal group received no treatment,BMT control group,granulocyte-colony stimulating factor(G-CSF)group,Shengxuezengbai group,and Shengxuezengbai+G-CSF group.BMT mouse models were established.Shengxuezengbai group and Shengxuezengbai+G-CSF group were given Shengxuezengbai Decoction.On d 9,14 and 24 after BMT,the mice were killed,and then peripheral blood cells and bone marrow nucleated cells were detected.The histological observation of bone marrow was made,and the expression of SCF was assayed by immunohistochemistry.Results:On d 14 after BMT,the level of hemoglobin in the peripheral blood cells in Shengxuezengbai group was higher than those in BMT control group and G-CSF group(P﹤0.05),and the level of hemoglobin in Shengxuezengbai+G-CSF group was higher than that in BMT control group(P﹤0.05).On d 24 after BMT,the level of white blood cells in the peripheral blood cells in Shengxuezengbai group was higher than that in BMT control group(P﹤0.05).In G-CSF group,Shengxuezengbai group and Shengxuezengbai+G-CSF group,the levels of bone marrow nucleated cells on d 24 after BMT were higher than that in BMT control group(P﹤0.01).The volume of hemopoietic tissues and the expression level of SCF in Shengxuezengbai group were higher than those in the control group on d 9 after BMT(P﹤0.01).Conclusion:Shengxuezengbai Decoction can enhance SCF expression in bone marrow after syngeneic BMT in mice,which may be related to the mechanism underlying Shengxuezengbai Decoction accelerating hematopoietic reconstitution.
