以上海某鸡场分离的1株对LoVo细胞及A549细胞具有良好的溶瘤效果且呈时间依赖性的新城疫病毒(Newcastle diseases virus,命名为SH15)为研究对象,依据Gen Bank中Mukteswar毒株,设计了10对引物以用于扩增SH15的片段cDNA,并对阳性克隆进行菌液PCR鉴定以及测序分析。测序分析比对结果确定,SH15株的全基因组长15 186 bp,在GenBank登录号为KY247177,结合生物学试验确定为中等毒力型。与其他毒株序列分析表明:该毒株与JS/9/05/Go、Mukteswar的全基因组同源性为99.8%,属于NDV的ClassⅡ分支中的基因Ⅲ型。推测SH15的溶瘤能力可能与HN蛋白中H203位点变异有关。本研究为了解新发现的溶瘤NDV毒株的生物学特性及全基因序列,以及以NDV为载体进一步开发肿瘤靶向药物奠定了基础。
The oncolytic strain of Newcastle disease virus(NDV)SH15 was used as material due to its good oncolytic effect on LoVo cell and A549 cell in a time-dependent manner. Ten pairs of primers were designed according to Mukteswar strain in GenBank and positive clones were screened by RT-PCR. Sequence analysis showed that the length of SHI5 was 15 186 nucleotides(GenBank accession No. KY247177),and pat- hogenicity tests indicated SHI5 belonged to Mesogenic strain. Phylogenetic analysis showed SH15 was classified as genotype III of Class II ,and its homology with JS/9/05/Go and Mukteswar strain was up to 99.8%.The H203 site of HNprotein may be the key to oncolytic effect of SHl5. The present study facilita- tes the further studies on the new isolated NDV strain,even development of antineoplastic targeted drugs based on NDV.
