胸腺素α1调节鸡脾脏淋巴细胞免疫功能的信号转导途径

在无菌条件下,从健康墟岗黄鸡体内取出脾脏,分离淋巴细胞并进行体外培养。在细胞培养液中分别加入终浓度为0.01mol/L咖啡碱、0.05mol/LEDTA、0.01mol/L氯化锂和5μg/mL甲派氟丙嗪,然后用10μg/mL胸腺素α1(Tα1)处理,再加入20μg/mL ConA,研究Ca^2+-肌醇磷脂-蛋白激酶系统和cAMP-肌醇磷脂-蛋白激酶系统在Tα1调节淋巴细胞免疫功能中的作用,揭示Tα1调节免疫功能的信号转导机制。结果表明,咖啡碱处理能显著增强Tα1促进脾脏淋巴细胞增殖转化、IL-2的基因表达,提高培养液中IL-2浓度(P〈0.05);而EDTA处理能显著抑制Tα1促进脾脏淋巴细胞增殖转化、IL-2的基因表达与分泌的作用(P〈0.05);氯化锂和甲派氟丙嗪对Tα1的免疫调节作用的影响不明显(P〉0.05)。结果提示,Tα1调节鸡脾脏淋巴细胞的增殖和IL-2的基因表达与分泌的作用,主要通过cAMP、Ca^2+和PKC等第二信使的介导。

英文摘要:

The signal transduction mechanisms of the regulation of thymosin α1 ( Tα1 ) on the immune functions of chicken splenic lymphocytes in vitro were studied. The lymphocytes were isolated from the spleen of XuGangHuang chicken in aseptic condition and cultured in vitro. The 0. 01 mol/L Caffeine, 0. 005 mol/L EDTA, 0.01mol/L LiCl and 5 μg/mL Amylozine were added into the culture medium, respectively. Then the lymphocytes were treated with 10 μg· mL^ -1 Tα1 and ConA was added to the culture medium. The action of Ca^2+ -inositol phospholipid-protein kinase system and the cAMP-inositol phospbolipid-protein kinase system during the regulation of Tα1 on lymphocyte immunity function was studied. The results showed that Caffein could promote proliferation of spenic lymphocyte and gene expression and secretion of IL-2 significantly ( P 〈 0. 05 ). EDTA inhibited the differentiation and proliferation of chicken splenic lymphocyte, the expression of IL-2 gene and the concentration of IL-2. LiCl and Amylozine had no significant effect on the immunity regulation by Tot1 ( P 〉 0. 05). The result implied that Tα1 significantly enhanced the immunological function of chicken splenic lymphocyte by the second messenger, such as cAMP, Ca^2+ , IP3 and PKC.
