目的:观察蝎毒多肽B5(SVPB5)对正常及辐射后的大鼠骨髓间充质干细胞(r BMSCs)生长的影响。方法:利用全骨髓细胞贴壁法分离纯化r BMSCs,体外扩增传代,取第3~5代细胞为实验对象。实验分为正常对照组(IR-control)、正常加药组(IR-SVPB5)、辐射对照组(IR+control)以及辐射加药组(IR+SVPB5)。分别进行细胞计数、衰老相关β半乳糖苷酶染色及集落形成单位计数。结果:SVPB5对正常及辐射r BMSCs均有促进增殖的作用,其中SVPB5处理3 d后,对正常r BMSCs开始便表现为明显的促增殖作用(P〈0.05),而对辐射r BMSCs则于第10、11天表现出明显的促增殖作用(P〈0.05)。SVPB5作用于正常r BMSCs后,其衰老染色阳性细胞率明显低于正常对照组(P〈0.05),其集落数明显高于正常对照组(P〈0.05)。结论:SVPB5能促进正常及辐射后r BMSCs增殖,增强r BMSCs的增殖潜能并缓解其衰老。
Objective: To observe scorpion venom peptide B5( SVPB5) on growth of normal and irradiated rat bone marrow-derived mesenchymal stem cells( r BMSCs). Methods: r BMSCs were separated and purified with adherent cultivation of whole bone marrow cells,then cultured and amplified in vitro. Passage 3-5 r BMSCs were used as experimental subjects,divided into normal control group( IR-control group),normal dosing group( IRSVPB5 group),irradiation control group( IR + control group) and the irradiation dosing group( IR + SVPB5group). Each group was counted for total cells,colony forming units and stained with senescence-associated β-galactosidase. Results: SVPB5 promoted proliferation of both normal and irradiated r BMSCs. Specifically,the proliferation-promoting effect became obvious as early as 3d of SVPB5 treatment in normal r BMSCs( P 〈 0. 05),which was not shown in irradiated r BMSCs until 10-11 d of the treatment( P 〈 0. 05). After SVPB5 treatment,the normal r BMSCs showed significantly lower percentage of senescence β-galactosidase staining positive cells and higher counts of colony forming units,compared with the normal control group( both P 〈 0. 05). Conclusion:SVPB5 may promote proliferation of normal and irradiated r BMSCs,enhance proliferation potential and mitigate cell aging of r BMSCs.
